Southern blotting is a detection technique used to find the target DNA sequences in the DNA sample in the field of molecular biology. Gene knockout has its application both in classical genetics and modern techniques such as functional genomics. During the initial timescale, gene knockout was carried n transposon mutagenesis. The advent of PCR meant that insufficiencies in the quantity of DNA were no longer a limitation in molecular biology research or diagnostic procedures. The chemistry involved in PCR depends on the complementarity (matching) of the nucleotide bases in the double-stranded DNA helix. When a molecule of DNA is sufficiently heated, the hydrogen bonds holding together the double helix are disrupted and the molecule separates or denatures into single strands.
