{"id":4418,"date":"2019-04-26T10:04:26","date_gmt":"2019-04-26T10:04:26","guid":{"rendered":"https:\/\/www.mybiosource.com\/learn\/?page_id=4418"},"modified":"2023-03-19T14:48:06","modified_gmt":"2023-03-19T14:48:06","slug":"alexander-disease-2","status":"publish","type":"page","link":"https:\/\/www.mybiosource.com\/learn\/alexander-disease-2\/","title":{"rendered":"Alexander Disease"},"content":{"rendered":"<p>Alexander disease is an extremely rare <span id=\"urn:enhancement-4dd8d6bf-b198-4388-9503-446181334215\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/neurological-condition\">neurological disorder<\/span> that occurs when the <span id=\"urn:enhancement-38ed57ab-fdcb-494c-b0a8-f30d30b74fc3\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/white-matter\">white matter<\/span> (<span id=\"urn:enhancement-811ee4c2-0328-47ce-b455-0de982ba9431\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/myelin\">myelin<\/span>) insulating the <span id=\"urn:enhancement-6821dcdf-7bde-48a0-988f-7cdec2ef978b\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/nerve-fibers\">nerve fibers<\/span> are destroyed.\u00a0 <span id=\"urn:enhancement-2f73e8ba-2a8e-4b87-9a6b-9f5fac0ae912\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/myelin\">Myelin<\/span> is essential for the effective transmission of signals between the nerves and when they are affected, the <span id=\"urn:enhancement-5780ffc7-94b6-4608-881a-7594b831f348\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/nerve-impulses\">nerve impulses<\/span> become disrupted.<\/p>\n<p>In individuals with alexander disease, the <span id=\"urn:enhancement-cefa0596-1dd3-4a0c-9dbb-e0328447ec9e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/white-matter\">white matter<\/span> is destroyed in the brain along with the abnormal protein deposits known as <span id=\"urn:enhancement-807bb007-30e4-4b7e-bc45-654ca8b7e0f2\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rosenthal-fibers\">Rosenthal fibers<\/span>. These fibers occur in the supporting cells of the brain known as <span id=\"urn:enhancement-96cd58bf-d6d9-4989-9f96-8183b2dade7b\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/astrocytes\">astrocytes<\/span>.<\/p>\n<p>Alexander\u2019s disease has been divided into three forms depending on the age of onset and their symptoms; infantile, juvenile and adult forms. The common feature of alexander disease is the abnormal protein accumulation in the various parts of the brain and the <span id=\"urn:enhancement-ef1680c1-e100-42b6-b9f1-735ef1bb6830\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/the-nervous-system\">central nervous system<\/span>. In the later onset of alexander disease, the <span id=\"urn:enhancement-4da7be93-a416-496f-aa4e-d4c231df1100\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/myelin\">myelin<\/span> defect may not be present. The infantile form of this disorder has a poor prognosis with most children not surviving past the age of 6. The juvenile and the adult onset tend to progress slowly.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>Causes <\/strong><\/p>\n<p>This disease can be inherited in an <span id=\"urn:enhancement-21d6e789-4b7f-4133-adcc-c305413a0b82\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/autosomal-dominant\">autosomal dominant<\/span> manner, where one copy of the defective <span id=\"urn:enhancement-6a63b961-8b11-40f1-8bcf-4a2e56605339\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/gene\">gene<\/span> causes the disorder. However, most cases of alexander disease are thought to be provoked as the result of a new genetic mutation that occurs spontaneously. \u00a0The genetic mutation identified to cause this condition is the <span id=\"urn:enhancement-022b8083-8e6c-4448-8280-8a9f82acbc61\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/gene\">gene<\/span> GFAP. This is responsible for the production of the <span id=\"urn:enhancement-49bc8cb7-db0c-4eed-a07c-1af2e565baf4\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/glial-cell\">glial<\/span> fibrillary acidic protein which attachs together to create <span id=\"urn:enhancement-8db8de90-b2f0-48b2-8ddd-87d69ec8a83b\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/intermediate-filaments\">intermediate filaments<\/span> necessary for the structural development of the cells. The defective <span id=\"urn:enhancement-ede3e829-6168-4577-918c-3cff9462fd47\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/gene\">gene<\/span> results in a structurally abnormal <span id=\"urn:enhancement-977302b1-b868-4781-9d41-06e6cb038f7d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/glial-cell\">glial<\/span> fibrillary acidic protein affecting the normal cellular function.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>Symptoms<\/strong><\/p>\n<p>The infantile form is usually presented with symptoms in the first few years of the infant\u2019s life. Although some may not survive past their first year, the others may continue to live for 5- 10 years of age. Severe <span id=\"urn:enhancement-546a6b6f-326d-442c-9ccf-aacda5148a9d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/mental-retardation\">mental retardation<\/span> occurs progressively and earlier the age of onset, the severe the condition progresses. Some of the symptoms of the infantile form include abnormally large brain, fluid accumulation in the brain, failure to grow or gain weight, seizures, difficulty with feeding, walking, speech and mental regression.<\/p>\n<p>Individuals with the juvenile form of alexander disease may have problems with swallowing and coughing. These individuals have a normal sized head and mental functions. As the disease progresses slowly, they may survive to middle age.<\/p>\n<p>An adult onset is a rare form characterized by impaired coordination, difficulties in speech, a problem with sleep and swallowing. The symptoms are often mild in this form of the disease and are similar to the juvenile form.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>Diagnosis<\/strong><\/p>\n<p>The <span id=\"urn:enhancement-f1e72ada-ec8f-4f26-bbea-7543dcd2e349\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/mri\">MRI<\/span> can deliver an accurate diagnosis of the typical early onset of the disease. Also, other medical conditions that present similar symptoms should be ruled out with various tests. A genetic testing can detect the defective <span id=\"urn:enhancement-dc78ab43-665b-4c12-8899-5c0fbd07b24e\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/gene\">gene<\/span> responsible for alexander disease. However, a small percentage of these cases may not be identified with the genetic mutation, hence the possibility of this condition cannot be ruled out.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>Treatment<\/strong><\/p>\n<p>There is no cure for this disease. Generally, the treatment focuses on relieving the symptoms associated with this condition. The other form of treatment includes supportive care.
