{"id":4423,"date":"2019-04-26T10:05:55","date_gmt":"2019-04-26T10:05:55","guid":{"rendered":"https:\/\/www.mybiosource.com\/learn\/?page_id=4423"},"modified":"2023-03-19T14:54:41","modified_gmt":"2023-03-19T14:54:41","slug":"allan-herndon-dudley-syndrome-2","status":"publish","type":"page","link":"https:\/\/www.mybiosource.com\/learn\/allan-herndon-dudley-syndrome-2\/","title":{"rendered":"Allan-Herndon-Dudley syndrome"},"content":{"rendered":"<p><strong><em>Allan-Herndon-Dudley syndrome<\/em><\/strong><\/p>\n<p>Allan-Herndon-Dudley syndrome is a rare <span id=\"urn:enhancement-aae957ea-054c-4676-9557-7bf5e214d52c\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/hereditary-disease\">inherited disorder<\/span> affecting the <span id=\"urn:enhancement-4ea46662-c54f-4bda-80a4-728f5f2c4d55\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/brain-development\">brain development<\/span>. This occurs mainly among the male population who exhibit moderate to severe <span id=\"urn:enhancement-a2052722-b9ec-4172-9e7b-34dfcefbe851\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/developmental-disabilities\">mental disabilities<\/span> and problems with their movements. Although the infants born with this condition appear normal, the symptoms become evident shortly after birth and include <span id=\"urn:enhancement-f210a76b-ad3f-4713-9a5c-9edb7b7e7d74\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/poor-muscle-tone\">poor muscle tone<\/span>, inability to control head movements and developmental delays. This rare condition has only been reported in about 25 families affecting 89 individuals globally.<\/p>\n<p><strong><em>Symptoms <\/em><\/strong><\/p>\n<p>The children affected with allan-herndon-dudley syndrome are presented with weak muscle tone (hypotonia) and problems with the muscles. As they become older, these individuals also develop joint deformities affecting the movements of joints. They may also exhibit muscle stiffness along with the involuntary movements of the limbs. In addition, other symptoms include poor growth, abnormal folding of the ears, developmental delay, poor head control and unclear or no speech. As these people are unable to walk on their own, they are often wheelchair users. Most of these individuals have survived into their 60s, however, the overall life expectancy is compromised.<\/p>\n<p><strong><em>Causes<\/em><\/strong><\/p>\n<p>This syndrome is inherited in an <span id=\"urn:enhancement-8e441182-245c-4402-aac9-e08893689345\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/x-linked-recessive\">X-linked recessive<\/span> pattern meaning the mutated gene is present in one of the two <span id=\"urn:enhancement-ccbef7b5-2196-4f76-85bb-0d6ab48472c4\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/male-hormones\">sex hormones<\/span>. The X-linked disorders only affect the male who carry one <span id=\"urn:enhancement-29f1a470-4af1-4047-b7c6-b992fd8bed6c\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/x-chromosome\">X chromosome<\/span> in comparison to the females who carry two. So, the single copy of the defective gene can cause the disorder, whereas the females will require both the copy of the gene to be defective to develop this condition. As this is more unlikely, males are more affected than the females who are often the carriers of the disorder. The X-linked disorders cannot be passed from the father to the son.<\/p>\n<p>The mutation in the gene SLC16A2 develops allan-herndon-dudley syndrome. This is responsible for delivering the instruction to produce a certain protein essential in the development of <span id=\"urn:enhancement-c55064d4-aec3-4a7d-948b-ac734fc31ac5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/the-nervous-system\">the nervous system<\/span>. This protein delivers a <span id=\"urn:enhancement-29d976f2-004b-40c4-a307-34f67d8ec5d8\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/hormone\">hormone<\/span> known as triiodothyronine (<span id=\"urn:enhancement-239d0ca1-b477-4201-8c07-d1c348b2ea65\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/t3\">T3<\/span>) into the <span id=\"urn:enhancement-7106fdaf-b8f3-445f-a4a7-d7c93af1cbc9\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/brain-cells\">nerve cells<\/span>. When the gene is defective, the function of the protein is affected causing problems in the transportation of <span id=\"urn:enhancement-c98bb460-fc77-44b5-a2b9-d45d753ff778\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/t3\">T3<\/span> <span id=\"urn:enhancement-d3a9100f-83c5-4481-a1d8-3d4239a86ac1\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/hormone\">hormone<\/span> to certain parts of the brain. This affects the normal development of the brain. As the <span id=\"urn:enhancement-b8752b3e-4514-4c21-a2e7-98b6b084bc3e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/t3\">T3<\/span> hormones cannot be effectively delivered, its accumulation affects different parts of the body causing symptoms associated with this condition.<\/p>\n<p><strong><em>Diagnosis<\/em><\/strong><\/p>\n<p>The diagnosis is based on the evaluation of clinical history and examination that shows a neurological and <span id=\"urn:enhancement-3efd29b6-7cb3-407e-b861-49f0ee623150\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/cognitive\">cognitive<\/span> decline. The laboratory blood analysis can identify elevated levels in free <span id=\"urn:enhancement-904bd5d6-75e0-400a-9cd9-00ee727ea6f6\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/t3\">T3<\/span> and decreased free <span id=\"urn:enhancement-ae326f72-c8ba-4dde-a46b-534b3d5d2768\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/t4\">T4<\/span>. The genetic testing can detect the mutation of the gene SLC16A2 (or MCT8) associated with this disorder.<\/p>\n<p><strong><em>Treatment<\/em><\/strong><\/p>\n<p>There is neither a cure nor an effective treatment available for allan-herndon-dudley syndrome.
