The <span id=\"urn:enhancement-ef62cdaf-1d25-4c90-99ed-45d0d4a0cf1d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergic\">allergic<\/span> reaction is stimulated by the proteins in the food product which causes the immunological reaction. Allergens are the compound which brings about the immunological response, the allergen is usually a heat sensitive water soluble protein within the range of 10-100 kDa with no commonality in structure or amino acid sequence having multiple <span id=\"urn:enhancement-7201b286-3901-493e-b36f-5ea17bc6e435\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ige\">IgE<\/span> <span id=\"urn:enhancement-486e3f07-3412-4688-8d6d-52ab510f2af0\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/binding-sites\">binding sites<\/span>. Modern food products are packed with various additives and preservatives which can cause <span id=\"urn:enhancement-8c144dd4-69da-413f-a2b0-b3e0af404808\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergic\">allergic<\/span> reactions in any individual. This prompts for screening of food products for potential allergens, in case of allergy causing compound presence in the food it has to be mentioned on the product label so that the individual can identify and avoid consuming the <span id=\"urn:enhancement-0d9d13d0-8a0d-43c4-a42d-a559f96bf39e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergenic\">allergenic<\/span> food. Strict avoidance needs to be followed by individuals with food allergies and food labels is used to indicate to these individuals the presence or absence of allergens.<\/p>\n<p>Due to the growing number of <span id=\"urn:enhancement-498ec78c-6c35-45a3-8e06-f8027704680b\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergic\">allergic<\/span> reactions, it is crucial to have fast, reliable methods of allergen detection in processed food products. Identification and labeling the <span id=\"urn:enhancement-8603e8c6-687b-4896-ad0f-10d48545d0a6\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergenic\">allergenic<\/span> food components is mandatory in various industrialized countries around the world. The presence of undeclared <span id=\"urn:enhancement-7b08f097-40ce-42e3-8cbe-e5698d552720\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergenic\">allergenic<\/span> food pose risk for those who suffer from food allergies. Some of the additives and processing aids used include proteins that includes important allergens such as milk proteins and egg white proteins. Milk and egg components have been used for the winemaking process as fining agents which according to the wine producer\u2019s knowledge, is removed after coagulation by filtration. Fining the wine aims to soften or reduce the astringency or the bitterness of the wine and it is also done to remove protein that can potentially form haze or in some cases it is added to stabilize and reduce the color by absorption and precipitation of polymeric phenolic compounds and tannins. Some of the common proteins used for fining are included in the list of allergens in the EU regulation that includes milk and egg proteins, animal <span id=\"urn:enhancement-1006513c-e5ca-4961-8b0d-b63680c71e72\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/gelatine\">gelatine<\/span>, fish <span id=\"urn:enhancement-e66ab172-e05d-4c49-a689-9bd5c1051028\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/gelatine\">gelatine<\/span>\/<span id=\"urn:enhancement-c94341a1-3c54-4f95-8b04-b97f3edbc56c\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/isinglass\">isinglass<\/span> and those derived from wheat and white lupin. The proteinaceous substances that are used as a fining agents during wine making, removes the unwanted insoluble particles and undissolved microscopic particles (colloidal material) from the must or wine in order to improve stability. The use of an egg allergen (<span id=\"urn:enhancement-f1440451-c5f0-486a-9de0-fdb464f69ef2\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/lysozyme\">lysozyme<\/span>) included as wine additives helps to control the <span id=\"urn:enhancement-9407bcf5-e584-4cff-b4d7-be790241d837\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/fermentation\">fermentation<\/span> processes and to avoid spoiling during winemaking. Even if the processing aids are filtered and removed in the final product, the trace elements must be excluded in order to avoid <span id=\"urn:enhancement-480e4c77-ff07-40e1-b978-6f1903696c41\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergic\">allergic<\/span> reaction to sensitized individuals. Hence, reliable detection and quantification methods for food allergens are important to ensure compliance with food labeling.<\/p>\n<p>Currently, the methods used to detect the potential allergens are the markers that indicates that presence of the allergens or the <span id=\"urn:enhancement-eb42d9c8-9985-4599-95ae-784733623cfa\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergenic\">allergenic<\/span> ingredient itself. In order to detect the allergens, the common <span id=\"urn:enhancement-a841a8bb-085f-4209-bd2d-79207685ef7a\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/analytical\">analytical<\/span> methods used include the immunochemical assays like <span id=\"urn:enhancement-e7e1eb9b-3f80-40fc-a7a3-0ddfd2d49421\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> (Enzyme Linked <span id=\"urn:enhancement-02f1f6b0-2205-4674-a2f2-d91ffd88f481\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/immunoassay\">Immunoassay<\/span>), DNA based method like <span id=\"urn:enhancement-5e32925d-4c20-4acf-8df9-404c50524d36\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/polymerase\">polymerase<\/span> chain reaction (<span id=\"urn:enhancement-3484d150-29af-4743-b29d-42d4a9951162\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/pcr\">PCR<\/span>) and <span id=\"urn:enhancement-a4a92063-266c-4d54-a9af-c835ae714a82\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/mass-spectrometry\">mass spectrometry<\/span>. The choice of allergen detection method depends upon the purpose, type of sample, food matrix, turn-around time, availability of equipment and cost<\/p>\n<p>Egg white proteins are one of the most common fining agents used for the red wines to remove phenolic compounds associated with the astrigiency in red wines. The main allergens of egg white are ovomucoid, <span id=\"urn:enhancement-130b3a57-09f6-4cf5-a1e4-6cd1312453f1\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ovalbumin\">ovalbumin<\/span>, ovotransferrin and <span id=\"urn:enhancement-d0c03ec9-a756-4058-8622-47c1bdb86310\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/lysozyme\">lysozyme<\/span>. The <span id=\"urn:enhancement-94b0f1ac-c15b-497c-b0bd-4ce5b0744652\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> test is the most important method used to detect the allergens because of its specificity and sensitivity and also because the equipment required is not expensive. \u00a0There are various <span id=\"urn:enhancement-1ae4cea2-769f-4230-848a-a5f2f8d10207\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> methods used to detect the presence of proteinaceous fining agent residues in wines and most of these assays are based on specific in-house antibody production which are difficult to implement elsewhere. Hence, commercially available kits are used by the laboratories for wine analysis and control.<\/p>\n<p><strong>ELISA Working and Principle<\/strong><\/p>\n<p><span id=\"urn:enhancement-91d1f2b7-8f1c-4a99-88d8-88d115025c32\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> is an antibody based detection method for the identification of protein or other protein in the food product. Three types of <span id=\"urn:enhancement-757c2bef-3351-456f-9768-f77ddac53e0a\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> test kit are available as well flow, lateral flow and sandwich format with sandwich method being the most commonly used technique for allergen detection in food industry. The analysis takes one-two hours to complete and it qualitative and quantitative test method. Lateral flow <span id=\"urn:enhancement-1c80699d-c5bb-458f-9f44-00ca30acad49\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> devices are qualitative and semi-quantitative with a reader available for many <span id=\"urn:enhancement-67e6200e-fe6c-4849-b6a7-9d412d300fa9\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergenic\">allergenic<\/span> food. The main advantages of <span id=\"urn:enhancement-606d7022-5f8b-49a7-a06e-e6af5628194f\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> is its sensitivity, it can detect for <span id=\"urn:enhancement-d40fe52e-1da3-44e7-9010-015a776b31e5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergenic\">allergenic<\/span> compounds in ppm range, it is fairly rapid compared to other methods, equipment needs are less and the kit requires low skill level to operate. Cross reactivity is the main disadvantage with <span id=\"urn:enhancement-92e61646-682d-481f-a267-1e0f39952a35\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> and the analyst needs to understand which kits to use as some milk kits detect <span id=\"urn:enhancement-60a2a42a-4a13-41cd-a92a-ccd2cfb82558\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/casein\">casein<\/span> while others detect whey proteins.<\/p>\n<p>Detection of allergens in food using <span id=\"urn:enhancement-5df4ca00-ce8e-46e9-9017-e52b922d1e72\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> requires the preparation of the microtitre plate well. <span id=\"urn:enhancement-bf593884-fb36-4833-853f-89655231586f\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/secondary-antibody\">Primary antibody<\/span> is immobilized on the microtitre plate well. To this 100 \u00b5l of sample containing the <span id=\"urn:enhancement-31110057-3b05-48b5-92b0-1060388837f1\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antigen\">antigen<\/span> is added and allowed for the <span id=\"urn:enhancement-2a569867-f79e-4c77-b424-2880d82dd24a\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/secondary-antibody\">primary antibody<\/span> and <span id=\"urn:enhancement-5d6e6c08-61da-4387-8a9c-790102a4deb8\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antigen\">antigen<\/span> from the sample to bind and form an antibody-<span id=\"urn:enhancement-f6df2581-031c-4ff2-b5bd-6825727f6455\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antigen\">antigen<\/span> complex. This is followed by a washing procedure to remove the unbound sample material. An enzyme conjugate is added to microtitre well containing the bound antibody-<span id=\"urn:enhancement-25ff3e3b-fd0c-4c6d-ba55-596459b3e5b0\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antigen\">antigen<\/span> complex, this results in the formation of antibody-<span id=\"urn:enhancement-23c0ee02-d642-46e6-a78b-8019045305c9\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antigen\">antigen<\/span>-antibody complex and to this a <span id=\"urn:enhancement-c917759a-9c06-46f3-bae7-2f1271458ebc\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/substrate\">substrate<\/span> or chromogen is added. The chromogen reacts with the enzyme conjugate to produce blue color, to which absorbance is then measured at 450 nm.\u00a0 The international organization of vine and wine (OIV) has also recommended the use of <span id=\"urn:enhancement-bfca72ce-ce7c-45f7-8eaa-6566eb4259fe\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> with a <span id=\"urn:enhancement-04c47209-159f-4cf4-8c7f-f307de3493a7\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/detection-limit\">limit of detection<\/span> (LOD) of &lt;0.25mg\/L and a limit of quantification (LOQ) of &lt;0.5 mg\/L (OIV 2014).<\/p>\n<p><strong>Multiplexed approach for detecting allergens using Immunochemical methods<\/strong><\/p>\n<p>Beads are labeled with antibodies for selected antigens like for whey or <span id=\"urn:enhancement-447b34af-db79-46a0-82a2-f178a3122773\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/casein\">casein<\/span>. Beads are then mixed and exposed to antigens for binding following which a second <span id=\"urn:enhancement-e0ac2f0b-b85f-4df9-8008-4f8fd0c8e370\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antigen\">antigen<\/span> specific labeled antibody is added for detection. The beads are then made to flow through an optical path of two lasers, where the first laser identifies bead by color and correlates it with specific <span id=\"urn:enhancement-b1c65421-242a-491f-9047-1c5735a867d1\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antigen\">antigen<\/span> while the second laser determines whether recognition event has occurred and how many times it has occurred. This test method can give results both in qualitative and quantitative.<\/p>\n<p><strong>PCR Based Analysis<\/strong><\/p>\n<p>Useful in situation when <span id=\"urn:enhancement-abe6b3b3-43c6-4c1d-be37-902cfcf909ee\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> is not applicable. Not practical for in-plant use as it involves expensive equipment and a dedicated lab. Also the test does not prove presence or absence of protein\/allergen Detects only <span id=\"urn:enhancement-fa407957-0a19-4100-ac96-bb2c181a8067\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dna-sequence\">DNA sequences<\/span> indicative of <span id=\"urn:enhancement-77921703-a180-497f-bbcd-0b72461b3dcb\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergenic\">allergenic<\/span> species and has no role in <span id=\"urn:enhancement-197ce22b-21e0-4394-a4de-27a503f5f4b7\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergenic\">allergenic<\/span> protein detection. The absence of DNA does not necessarily indicate absence of proteins as well. The equipment is costly and requires skilled personnel. In protein enriched isolates or concentrates DNA has been proven to be an alternative molecular marker for the presence of an <span id=\"urn:enhancement-2d084aaa-f726-4c58-8c0f-02114f802968\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergenic\">allergenic<\/span> food. This method is based on heat stable DNA <span id=\"urn:enhancement-de1f8533-9b93-4675-b542-e24cc45a751e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/polymerase\">polymerase<\/span> amplifies DNA fragment. Methods applying detection of <span id=\"urn:enhancement-991de83a-737f-4a43-abb8-2b8b930be0be\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/coding-sequences\">coding sequences<\/span> of <span id=\"urn:enhancement-93291b5c-294d-421a-879f-3958a167e7a4\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergenic\">allergenic<\/span> proteins allow direct analysis of <span id=\"urn:enhancement-54460140-95fa-4c34-8522-e671f2de1d91\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/allergenic\">allergenic<\/span> food components.
