Like all viruses, the <em><span id=\"urn:enhancement-adb18264-2e98-4abc-8c61-ed5d8c024f09\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/flavivirus\">flavivirus<\/span> <\/em>requires a host for its replication &amp; survival and it is able to transmit the disease to humans by using the mosquitoes, <em><span id=\"urn:enhancement-ddd8b07a-0d9c-4d09-a7f4-dbe4d95a2531\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/aedes-aegypti\">aedes aegypti<\/span> <\/em>as a <span id=\"urn:enhancement-4a14f382-65c9-408d-8fa6-830ae4284aec\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/vector\">vector<\/span>. <span id=\"urn:enhancement-0f0b35f8-5aeb-40b2-b5bc-bc20b536a320\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dengue\">Dengue<\/span> is one of the most prevalent disease caused by mosquitoes. In order to continue its life cycle the <em><span id=\"urn:enhancement-137c867f-f921-4db5-bca0-dec82c5d27ee\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/flavivirus\">flavivirus<\/span><\/em> needs to be transported using a <span id=\"urn:enhancement-2fe92c48-79e1-419c-9e24-ebadbfa8d7d6\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/vector\">vector<\/span>, recent research findings revealed that during the infectious stage in humans, the <em><span id=\"urn:enhancement-d1f8f9e3-c749-44a3-ab70-a78c34665686\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/flavivirus\">flavivirus<\/span><\/em> causes the infected patients skin to release certain <span id=\"urn:enhancement-dd6ea396-2320-4942-bbd7-f8a0a1caf771\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/volatile-organic-compounds\">volatile organic compounds<\/span> which further attracts more mosquitoes and makes the patient prone to mosquito bites, now these mosquitoes will further act as carrier and will infect the more people leading to a pandemic in the region. There are four main <span id=\"urn:enhancement-90a7cf71-aa4f-4559-b60e-ce68bcfcdcff\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/serotypes\">serotypes<\/span> of <span id=\"urn:enhancement-fe8486e2-90c3-4ec6-a005-ef06d778fe1d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dengue\">dengue<\/span> virus, they are DENVI, DENV2, DENV3 and DENV4 with some being specific to particular a region of the globe. When a mosquito (<em><span id=\"urn:enhancement-12298a35-29c7-46cc-90c3-76fd11316537\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/aedes-aegypti\">aedes aegypti<\/span>) <\/em>carrying the virus bites a human being, the virus enters the blood stream and starts to replicate in the liver for a period of 2-5 days and results in the lysis of <span id=\"urn:enhancement-dcc7b19d-5ee5-4d85-a67a-4996c724e5b1\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/hepatocytes\">hepatocytes<\/span> releasing more virus which will further infect more cell and multiply. During this phase the virus destroys the platelets resulting in <span id=\"urn:enhancement-52657c7a-0efb-44c0-977d-11539e980d52\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/thrombocytopenia\">thrombocytopenia<\/span>, a condition where the platelets are below the normal level of 1,50,000\/cu mm. With less platelets the body is unable to stop internal bleedings resulting in <span id=\"urn:enhancement-882f293f-bf74-496e-8058-b4c9212ebf44\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/hemorrhagic\">hemorrhagic<\/span> fever and will lead to death of the patient. Current treatment options to increase the <span id=\"urn:enhancement-04179530-4b40-410b-8d63-bdfd8188bfa7\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/platelet\">platelet<\/span> levels are thrombopoietin <span id=\"urn:enhancement-533ff030-ed3b-49e1-87da-d005f7b1b31e\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/hormone\">hormone<\/span> mimetic, <span id=\"urn:enhancement-fbbb1d54-60e7-4cbc-ab5c-97b8e09d6cca\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/platelet\">platelet<\/span> transfusion and carica papaya leaf extracts. <span id=\"urn:enhancement-7572e550-e1b9-4e27-ba84-2b9b19316eff\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/platelet\">Platelet<\/span> transfusion is usually performed as the last option as in some cases the <span id=\"urn:enhancement-63ef1a59-e8e5-481b-9b0d-5b707f2f980f\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/platelet\">platelet<\/span> transfusion can cause fluid buildup and this itself can lead to the death of the patient. Also, there have be no <span id=\"urn:enhancement-a0e56f15-66bb-4e13-8abd-47093c67ea60\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/clinical-trial\">clinical trials<\/span> reported to have been initiated for any antiviral activity of <span id=\"urn:enhancement-7ee424fd-c49d-48f2-86d0-7151e590bb78\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dengue\">dengue<\/span> so far.\u00a0 Considering the current scenario the global pandemic threat posed by this viral disease is a major cause of concern and more research is required to screen for an effective antiviral drug. Various phyto chemicals and algal biocompounds have exhibited potential antiviral activity for all four <span id=\"urn:enhancement-9ee4ed5e-8610-4586-b92b-2754ef57790d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/serotypes\">serotypes<\/span> of <span id=\"urn:enhancement-f6dc5a6d-9936-4b01-b1f7-b291f50411cd\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dengue\">dengue<\/span> virus. There are various methods that have been established to screen for antiviral activity, each has its own advantages and limitation. The main limitation of these methods are time in sample collection, transportation and sample pre-preparation. The immunodetection technique using <span id=\"urn:enhancement-b2bae659-63e1-45bb-a6c3-3d9b32af9a41\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> can be performed on the site to test for these antiviral activity saving time and allowing more potential samples to be screened for its antiviral activity.<\/p>\n<p><span id=\"urn:enhancement-d571d4f5-6a22-4a2d-8fcd-5c5d3d09c086\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span>, is defines as enzyme-linked immunosorbent assay and also called as EIA, it is a test that detects and measures <span id=\"urn:enhancement-4aad4a8c-c941-429f-819b-47e247c7bf4c\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dengue\">dengue<\/span> antibodies like <span id=\"urn:enhancement-61a56196-2f2a-40cc-99c5-c1d9f8c0e12c\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ns1\">NS1<\/span>, <span id=\"urn:enhancement-86e81b23-4188-4442-a35f-09c047f00ea8\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/igg4\">IgG<\/span> and <span id=\"urn:enhancement-6275aa4f-d928-4852-bb3b-b77b99214bd9\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/igm\">IgM<\/span> in one\u2019s blood. These antibodies are proteins that the body produces in response to antigens. After 3 days for assay validation wells are infected with DEN V1, DENV2, DENV3, and DENV4 at a multiplicity of infection ranging from 0.01 \u2013 4. A second validation which will act as control and it is done by infecting the wells with DENV4 at a lower multiplicity of infection (MOI) followed by treatment with <span id=\"urn:enhancement-379c841d-2626-48f3-8d68-d29b83843d1d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ifn-%ce%b1\">IFN-\u03b1<\/span> 2A. <span id=\"urn:enhancement-4b5a54b1-f53c-4472-865a-2e8bf024c18b\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ifn-%ce%b1\">IFN- \u03b1<\/span> is an antiviral drug used in the treatment of <span id=\"urn:enhancement-72c10537-3243-48bd-a2d3-c8d374a376c5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/hepatitis-c\">hepatitis C<\/span> infection, and proved to be an effective in vitro replication inhibitor of several <span id=\"urn:enhancement-21ea127c-fced-4a4a-bea2-135e6b07d3db\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/pathogen\">pathogenic<\/span> <em><span id=\"urn:enhancement-ced04b1c-4098-4546-a3de-89dd2874f64e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/flavivirus\">flavivirus<\/span><\/em>, including <span id=\"urn:enhancement-c51fae93-4bad-4169-9259-883a9df27975\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dengue\">dengue<\/span>. Based on that, <span id=\"urn:enhancement-da8d63fb-763d-4189-a4ec-4a921fcad71b\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/interferons\">interferon<\/span>-a 2A is used as a reference control. The results of the foci-forming assay, the <span id=\"urn:enhancement-d85667a3-97c3-4a6e-8215-838b5de3f911\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/in-situ\">in situ<\/span> <span id=\"urn:enhancement-ffb9d40a-e020-427e-b9b6-54d100af79fe\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> and a commercially available <span id=\"urn:enhancement-32409fd3-7f6b-44b4-8e61-a56122fb08f3\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ns1\">NS1<\/span>, <span id=\"urn:enhancement-ecd00e20-f370-4976-84e6-bf0b0e69f684\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/igg4\">IgG<\/span>, <span id=\"urn:enhancement-5bad184b-c555-41c6-8fe2-5d5c21b520a4\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/igm\">IgM<\/span> <span id=\"urn:enhancement-60c5c0be-a882-4438-9dbe-5dbeb6cfc285\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antigen\">antigen<\/span> <span id=\"urn:enhancement-0d42feea-9b77-4777-8f52-85c4d2eb7b32\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> were compared. <span id=\"urn:enhancement-1e574c4b-7b2a-4663-8e8c-aba0fda47dbe\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ifn-%ce%b1\">IFN-\u03b1<\/span> 2A was then used as a reference control and the <span id=\"urn:enhancement-620e39f4-f111-4022-a036-5773e99ab292\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ic50\">IC50<\/span> (concentration that inhibits 50% of virus infection) was determined using a dose response curve for all <span id=\"urn:enhancement-3b51cc0d-9cf9-43e5-ae3c-777308bec149\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dengue\">dengue<\/span> virus <span id=\"urn:enhancement-5c87db68-38b8-49b4-be8a-7bcac8f1e938\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/serotypes\">serotypes<\/span>.<\/p>\n<p>With the preparation of control and determination of concentration that inhibits the virus infection, the antiviral activity of any plant or marine seaweed of interest can be determined. The marine seaweed \/ plant sample of interest is screened and a negative and positive control is prepared in the test plates. The <span id=\"urn:enhancement-da80c461-f09c-4aea-90c2-268061c2b4c1\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> kit wells is then added with heptoma cells at a density of 2 X 10<sup>4 <\/sup>and then infected with different <span id=\"urn:enhancement-6f5b089f-7eb5-42a2-861d-94ade61bd3a5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/serotypes\">serotypes<\/span> of <span id=\"urn:enhancement-8de4c4cb-2f51-46bb-8327-b4814d386ebc\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dengue\">dengue<\/span> virus at a multiplicity of infection of 4.0, the plates are then incubated for 2 hours at room temperature. The inoculum is then replaced with 200 ml of plant or seaweed extract dilution. After a 72 h incubation period, cells were fixed and <span id=\"urn:enhancement-e1dda3f6-e371-40c4-8478-ee982ee32ada\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> is performed as previously described. Data were normalized as % of infection in relation to the controls, where the OD obtained with the non-infected culture were taken as 0% infection, and the one obtained with the non-treated infected culture as 100% infection.<\/p>\n<p>A disadvantage of this method is that <span id=\"urn:enhancement-2b76229a-77bf-4ff6-9535-39525a270fff\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/cytotoxic\">cytotoxicity<\/span> of the antiviral compound cannot be determined by this method, and hence it is advised to perform a cell viable test before screening for antiviral activity using <span id=\"urn:enhancement-b3479fd9-4741-41d4-ac29-c7dbd30e75c2\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/elisa\">ELISA<\/span> method.
