So, what is the buzz about? In this post, the aim is to discuss the <a href=\"https:\/\/www.broadinstitute.org\/what-broad\/areas-focus\/project-spotlight\/crispr-timeline\" target=\"_blank\" rel=\"nofollow noopener\">seminal works <\/a>briefly and where <span id=\"urn:enhancement-1448200d-9b4d-4cb3-80de-93d4d9ec604f\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/crispr\">CRISPR<\/span> would find its application.<img loading=\"lazy\" decoding=\"async\" class=\"alignright size-medium wp-image-238\" src=\"https:\/\/www.mybiosource.com\/learn\/wp-content\/uploads\/2017\/03\/crispr-genome-genetic-modification-dna-300x300.jpg\" alt=\"\" width=\"300\" height=\"300\" srcset=\"https:\/\/www.mybiosource.com\/learn\/wp-content\/uploads\/2017\/03\/crispr-genome-genetic-modification-dna-300x300.jpg 300w, https:\/\/www.mybiosource.com\/learn\/wp-content\/uploads\/2017\/03\/crispr-genome-genetic-modification-dna-150x150.jpg 150w, https:\/\/www.mybiosource.com\/learn\/wp-content\/uploads\/2017\/03\/crispr-genome-genetic-modification-dna-370x371.jpg 370w, https:\/\/www.mybiosource.com\/learn\/wp-content\/uploads\/2017\/03\/crispr-genome-genetic-modification-dna-570x571.jpg 570w, https:\/\/www.mybiosource.com\/learn\/wp-content\/uploads\/2017\/03\/crispr-genome-genetic-modification-dna-579x580.jpg 579w, https:\/\/www.mybiosource.com\/learn\/wp-content\/uploads\/2017\/03\/crispr-genome-genetic-modification-dna.jpg 660w\" sizes=\"(max-width: 300px) 100vw, 300px\" \/><\/p>\n<p>The years between 1993 and 2005 can be recognized as the discovery phase of <span id=\"urn:enhancement-1bb46741-06b4-409a-8c16-4a92bc3d7b01\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/crispr\">CRISPR<\/span>. \u00a0Francisco Mojica had been working on the locus (disparate repeat <span id=\"urn:enhancement-3eff9662-7f66-45ee-b7cb-dd9588687ea0\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dna-sequence\">sequences<\/span>) for almost two decades which is now the hallmarks of <span id=\"urn:enhancement-4dda462d-098b-43d3-95c5-a0770c1c3d97\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/crispr\">CRISPR<\/span> <span id=\"urn:enhancement-80a222e8-bb8b-4404-91c8-d3dbf5adb440\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dna-sequence\">sequences<\/span>. The term <span id=\"urn:enhancement-f02bb12c-d21f-4541-a069-141ebd958ad6\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/crispr\">CRISPR<\/span> was jointly coined by Ruud Jansen and Francisco Mojica.<\/p>\n<p>In May 2005, Alexander Bolotin of French National Institute for Agricultural Research (INRA) studying Streptococcus thermophiles discovered an unusual <span id=\"urn:enhancement-4e312132-7e87-4f19-a3ad-7839155b14e5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/crispr\">CRISPR<\/span> locus, the prime difference being novel CAS genes. One of the CAS genes now known as CAS9 that codes for protein is predicted to have nuclease activity. In addition, the spacers were identical to that of viral genes, also sharing common sequence in the end. This sequence, Protospacer adjacent <span id=\"urn:enhancement-a27ee3c3-8daf-49b5-a9a9-7f6f7f0f55b4\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/motif\">motif<\/span> (PAM) is required for target recognition.<\/p>\n<p>A notable contribution came from Luciano Marraffini and Erik Sontheimer of Northwestern University who demonstrated the target molecule is in fact \u00a0<span id=\"urn:enhancement-fe571179-7bd5-41fc-ad3d-ec0a85980994\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dna-strand\">DNA<\/span> and not <span id=\"urn:enhancement-0da72827-bcc9-44ca-bc43-8eda0963721f\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span>. The authors explicitly observed in the paper that the system could be powerful tool when transferred to non-<span id=\"urn:enhancement-6085d68c-fed3-4528-afb4-9f43a0243049\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/bacterial\">bacterial<\/span> systems. However, different type of <span id=\"urn:enhancement-9dfad78f-152a-44b8-9025-680f00946ad1\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/crispr\">CRISPR<\/span> system that target <span id=\"urn:enhancement-406105dd-28bd-4b90-aada-8fd8c9c2dacf\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> was also identified.<\/p>\n<p>The further advancements such as <span id=\"urn:enhancement-ce1a59ca-93aa-4879-bdaf-7b17da9081e8\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/cas9\">Cas9<\/span> ability to cleave target <span id=\"urn:enhancement-039af5a2-6e37-443a-9d05-5f79c9e672c0\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dna-strand\">DNA<\/span>, discovery of tracrRNA for <span id=\"urn:enhancement-5a2f1d1f-2528-41fd-9634-6f6f5ad20628\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/cas9\">Cas9<\/span> system, <span id=\"urn:enhancement-08f96549-ddac-4e3d-8458-48b2c6a0404b\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/crispr\">CRISPR<\/span> systems\u2019 ability to function heterologously in other species and biochemical characterization of <span id=\"urn:enhancement-459fc438-9730-49c6-b073-eb146a1867d6\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/cas9\">Cas9<\/span>-mediated cleavage helped scientists to rely on <span id=\"urn:enhancement-3d6cfd21-1e65-4b5a-9cc7-8abd27674414\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/crispr\">CRISPR<\/span> as <a href=\"https:\/\/www.mybiosource.com\/learn\/gene\/\">gene<\/a> editing tool for the future in the world of <span id=\"urn:enhancement-8b80d6d6-531d-47cf-a0fa-c63ba07a078e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/genome-engineering\">genome engineering<\/span>.<\/p>\n<p><span id=\"urn:enhancement-63c0564e-5045-41b8-bf4d-09555ba6a1f4\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/crispr\">CRISPR<\/span> system is viewed as a viable tool to create molecular pharmacies, destroy <span id=\"urn:enhancement-fe316d09-b0a9-4a8d-99b7-565f144398c0\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/viruses\">viruses<\/span> such as <span id=\"urn:enhancement-18646ac2-ef12-4205-9151-7aaba9d7874c\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/hiv\">HIV<\/span>, <span id=\"urn:enhancement-18748ead-ad62-4dac-bf14-7ff7bf200b01\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/herpes\">Herpes<\/span> and <a href=\"https:\/\/www.mybiosource.com\/learn\/hepatitis\/\">Hepatitis<\/a>, <span id=\"urn:enhancement-5d40fbd9-6488-46bc-b075-58275b5a2e8e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/crispr\">CRISPR<\/span> modified plants to reduce dependence on petrochemicals, precision system \u00a0to develop new cancer treatments and most importantly study <a href=\"https:\/\/www.mybiosource.com\/learn\/diseases\">diseases<\/a> such as like Alzheimer\u2019s and <span id=\"urn:enhancement-667216b5-e93a-4aa3-91d1-f632395fa63e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/parkinsons\">Parkinson\u2019s<\/span>.<\/p>\n<p>Plant biologists also see <span id=\"urn:enhancement-fba4eac1-2d01-4eef-be60-abb3973d5812\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/crispr\">CRISPR<\/span> as a viable system to enhance plant\u2019s ability thereby food security and figure out the functions of genes in complex environments.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Table of Contents I.
