It is estimated that around 75% of CRC cases are sporadic suggesting environmental and genetic as contributing factors while 25% is the result of inheritance. Most of CRC develop on the inner lining of the colon or <span id=\"urn:enhancement-128d6771-c7f0-4227-b749-a28db51d9a5d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rectum\">rectum<\/span> starting as <span id=\"urn:enhancement-3bcc4fab-e302-4c74-acc4-1db62dd55179\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/polyps\">polyps<\/span> and the chance of it developing into cancer is based on the type. The two main types of <span id=\"urn:enhancement-c6d49a0e-0585-4335-a54f-3602cca3e21c\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/polyps\">polyps<\/span> are; <span id=\"urn:enhancement-c851b70a-9811-4d35-87df-0a3c0f06763b\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/adenoma\">adenomatous<\/span> <span id=\"urn:enhancement-333590ce-210a-4b9e-a4a0-8a2ac39b2a8c\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/polyps\">polyps<\/span> also known as a <span id=\"urn:enhancement-c1d8cf36-2ffc-4526-8441-bb25581f5238\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/pre-cancerous\">precancerous<\/span> condition with the potential to develop into cancer and hyperplastic <span id=\"urn:enhancement-0b8ddd42-a06f-4be0-8834-6233b8e2209e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/polyps\">polyps<\/span> and inflammatory <span id=\"urn:enhancement-1355ed1e-e676-41f8-a37b-087acfbc6ee5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/polyps\">polyps<\/span> that are more common but are not considered <span id=\"urn:enhancement-a73c2392-04af-4813-a27d-21b83a6f4779\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/pre-cancerous\">precancerous<\/span>. Some of the lesser common types of <span id=\"urn:enhancement-f37aee0a-d08a-4826-bf9c-4560bc7ae4df\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> <span id=\"urn:enhancement-d3d58a96-c5a9-4bc8-8b90-5ae8db56f3f6\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/invasive\">cancers<\/span> include <span id=\"urn:enhancement-fa5acdd4-836f-44c0-a44c-508c872f44f0\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/carcinoid\">carcinoid<\/span> tumors, <span id=\"urn:enhancement-cf2cc932-5ed4-4df0-95e2-327eac215bef\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/gastrointestinal\">gastrointestinal<\/span> stromal tumors, <span id=\"urn:enhancement-bb00d958-c59d-4314-ad3d-ab1aa21af706\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/lymphoma\">lymphomas<\/span> and <span id=\"urn:enhancement-ea78fbfc-ec5a-4dc6-a155-2a47fc6d7908\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/sarcomas\">sarcomas<\/span>. In a majority of the cases, cancer remains undiagnosed until it has reached an advanced stage. Although the overall incidence of <span id=\"urn:enhancement-0aa791ed-c803-4db2-8ba2-fff3295bd2e3\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer for individuals over the age of 50 has decreased, according to the American Cancer Society, the trend of individuals younger than 50 years developing <span id=\"urn:enhancement-5d576163-90e1-4e82-9c50-f745d8123e40\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer has risen. Therefore, the American Cancer Society has now revised the guideline for screening for <span id=\"urn:enhancement-0f047fb0-fc1c-4a89-bff9-9c0d0bb99f91\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer at 45 years of age. As <span id=\"urn:enhancement-76af3fc7-23b9-4724-9490-f80069c64a17\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer may not present any symptoms in the initial stage of the disease, the visual examination of the stool is important to detect the abnormality. Some of the symptoms associated with CRC include <span id=\"urn:enhancement-07756fdd-96be-4532-8365-9fb3727756ea\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rectal-bleeding\">rectal bleeding<\/span>, blood in the stool, changes in bowel habits such as constipation or diarrhea, abdominal pain, fatigue and weight loss. Although the current availability of <span id=\"urn:enhancement-2e8bb2be-7648-41fe-ab5f-b62db0f307df\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antitumor\">chemotherapies<\/span> and biologics has increased the survival of the patients, research is continuing to identify novel treatments to target the tumor.<\/p>\n<p id=\"2\">According to a recent study conducted by the Indian Institute of Science, Education and Research, <span id=\"urn:enhancement-8387499f-c7ce-4970-8eb4-a17e12c86434\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer can be detected with a simple test devised to identify the abnormal imbalance in the special AT-rich binding <span id=\"urn:enhancement-08dc81c1-0cbc-49ff-808b-d986f5c3c497\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/proteins\">proteins<\/span>. This can help with the early diagnosis of cancer and to help initiate an early treatment.<\/p>\n<p><em>\u2018It is for the first time we found that any variation observed in the two types \u2013 SATB1 and SATB2 protein levels reported in cancer patients \u2013 can actually indicate the stage of <span id=\"urn:enhancement-80120e32-0d3b-4fd1-bed1-2f2dc84d3340\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer. Importantly, it can provide an estimation of the survival of patients,\u201d Sanjeev Salande, lead investigator of the study told media.<\/em><\/p>\n<p><strong><span id=\"urn:enhancement-3d4c4223-6a43-43d9-9893-047b2113c580\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/microsatellite\">Microsatellite<\/span> Instability (<span id=\"urn:enhancement-c39e31e4-ad83-447d-b246-a489154fcb7a\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/msi\">MSI<\/span>)<\/strong><\/p>\n<p id=\"3\">Microsatellites are the repeated stretches of DNA of the human genome and are prone to the high mutation rate. <span id=\"urn:enhancement-57122ff5-4380-4b1c-ab3b-132cdb3e854f\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/microsatellite\">Microsatellite<\/span> instability occurs because of a defective <span id=\"urn:enhancement-ef44a1cc-e12f-4f20-9fb2-7dc958ca9f5d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dna-mismatch-repair\">DNA mismatch repair<\/span> system and it is used diagnostically for the tumor identification and classification. As <span id=\"urn:enhancement-01a70650-76b7-4c73-aba9-2071e5d63381\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/msi\">MSI<\/span> is a genetic marker for CRC which can be used for prognosis and the prediction of which <span id=\"urn:enhancement-e4ad58f2-b3aa-46ab-b715-7be35e9217e6\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antitumor\">chemotherapeutic<\/span> treatment works, it can be detected indirectly by immunohistochemical (IHC) staining and by PCR-based amplification of specific <span id=\"urn:enhancement-34596656-c20c-4728-847a-0aaf157f2946\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/microsatellite\">microsatellite<\/span> repeats. Also, the <span id=\"urn:enhancement-073775bc-a7e9-4ec8-9b78-557db48e49ea\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> tumors with <span id=\"urn:enhancement-816a903e-bd06-4e75-ae8d-5d8e399e7ceb\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/msi\">MSI<\/span> tend to have a better prognosis than the other MSS forms of tumors. The three <span id=\"urn:enhancement-27246051-cf07-4a6b-a600-e143e3c18acf\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antitumor\">chemotherapeutics<\/span> used for CRC treatment include antimetabolites, alkylating agents and topoisomerase inhibitors.<\/p>\n<p>&nbsp;<\/p>\n<p id=\"4\">Colorectal Cancer Screening Guidelines from the American Cancer Society<\/p>\n<p>In the United States, <span id=\"urn:enhancement-6cc5a358-8761-4dd5-beee-a94279f9edbe\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer is the second leading cause of death from cancer. The screening is associated with a significant reduction in the incidence of <span id=\"urn:enhancement-993cd545-8530-466c-a09f-e1c833a67a6d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer and with the timely removal of <span id=\"urn:enhancement-75887743-a9c1-4b18-b2e5-8d6b34c725fe\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/pre-cancerous\">precancerous<\/span> lesions. According to the recommendation, adults aged 45 and above with the risk of CRC should undergo regular screening either by a visual examination or a high-sensitivity stool-based test and any positive result should be followed up with a <span id=\"urn:enhancement-4fa1045f-c8c0-45e4-9818-c856e62e3559\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colonoscopy\">colonoscopy<\/span>. However, a strong recommendation is suggested for individuals over the age of 50 and above and for those aged between 76 and 85, screening should be based on the individual\u2019s preference, health status and prior screening history. The screening options include fecal immunochemical test annually; multitarget stool DNA test every 3 years, computed tomography colonography every 5 years and flexible <span id=\"urn:enhancement-4c102142-209b-4311-af08-594f76055397\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/sigmoidoscopy\">sigmoidoscopy<\/span> every 5 years.<\/p>\n<p>&nbsp;<\/p>\n<p id=\"5\">Advancing <span id=\"urn:enhancement-fb84fd54-54ae-47e5-bfc6-268d7b6d50ad\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">Colorectal<\/span> Cancer Immunotherapies<\/p>\n<p>With immunotherapy becoming the standard of treatment for several <span id=\"urn:enhancement-14eec93a-2754-42e4-a329-d75deb2dcda5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/invasive\">cancers<\/span>, <span id=\"urn:enhancement-17a1ce31-141c-41f1-9ada-eb4ccdfacf5a\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer has remained resistant and its efficacy has remained limited with a small percentage of patients with <span id=\"urn:enhancement-3b7e053b-7132-45b8-a021-b099d6bf89db\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/microsatellite\">microsatellite<\/span> instability-high. The <span id=\"urn:enhancement-ccd4f355-b118-4777-90f6-47f56268b4a5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/cancer-research\">Cancer Research<\/span> Institute and Fight <span id=\"urn:enhancement-c394d729-3340-4f1d-83b4-9adbdb2a928d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">Colorectal<\/span> Cancer convened to develop a blueprint for research and guideline development for the advancement of immunotherapy as treatment of <span id=\"urn:enhancement-c0594291-233a-442b-8186-875bec27984e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer. In a clinical phase II trial, the use of pembrolizumab resulted in the stability of the disease among <span id=\"urn:enhancement-2d375c43-4836-487f-b386-52bbaa16a7d5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/msi\">MSI<\/span>-H patients which has resulted in the approval for pembrolizumab in all MSH-H cancer. Also, the advances in the taxonomy of <span id=\"urn:enhancement-caafea2f-3c4e-473c-ab85-68050ee6275e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer have resulted in the understanding of the pathways that are potential immunotherapeutic targets. Various studies investigated radiation or <span id=\"urn:enhancement-c1d8b0dc-9981-4708-b58b-9321abbc30ef\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/antitumor\">chemotherapy<\/span> in <span id=\"urn:enhancement-1bf88aef-76d4-4d78-aaf4-7e47ce62b848\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer and some explored the effects of combining them with immunotherapy. However, several questions still remain which may be answered by performing deep immune phenotyping. Further studies are required that analyzes the genomic profiles of the patients which could help understand why some individuals respond well to immunotherapy while others don\u2019t.<\/p>\n<p>&nbsp;<\/p>\n<p id=\"6\">New Treatments approved by <span id=\"urn:enhancement-190f0b18-1e5d-4883-adfd-78adb02cec2c\" class=\"textannotation disambiguated wl-organization\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/food-and-drug-administration\">FDA<\/span><\/p>\n<p><span id=\"urn:enhancement-2115c341-3084-4426-b0dd-881e6b4870e3\" class=\"textannotation disambiguated wl-organization\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/food-and-drug-administration\">FDA<\/span> recently approved the use of a combination of immunotherapeutics such as ipilimumab (Yervoy) and nivolumab (Opdivo) for certain individuals with <span id=\"urn:enhancement-c6dfadf3-8c56-4609-87a0-f46922e64bad\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer. The <span id=\"urn:enhancement-157a07bf-9a47-4fa3-8631-070963435ec0\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/microsatellite\">microsatellite<\/span> instability-high <span id=\"urn:enhancement-2ec22e64-110d-4c64-af10-5babd4d543a1\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer has shown responsive to the immunotherapeutic treatment by removing the brakes off <span id=\"urn:enhancement-3df5a558-2825-4572-bc25-81726434159c\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/immune-cells\">immune cells<\/span> called T cells. In May 2017, <span id=\"urn:enhancement-f69ae2dd-9753-4d42-af0e-9bbcdbfaf910\" class=\"textannotation disambiguated wl-organization\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/food-and-drug-administration\">FDA<\/span> approved the use of pembrolizumab for the treatment of <span id=\"urn:enhancement-f23cef4c-0b5c-4cbf-b9ca-2414b5b3d815\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/metastasized\">metastatic<\/span> solid tumor identified to be <span id=\"urn:enhancement-ff13c700-9dd2-40a6-ad6d-e54d807fa8db\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/microsatellite\">microsatellite<\/span> instability-high or mismatch repair-deficient <span id=\"urn:enhancement-529fb426-6f17-4946-8ef0-1af052e27e64\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/colorectal\">colorectal<\/span> cancer and in August 2017, <span id=\"urn:enhancement-641de49e-3f7b-43b9-8e03-54894c770da5\" class=\"textannotation disambiguated wl-organization\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/food-and-drug-administration\">FDA<\/span> approved nivolumab also.
