The costs to our healthcare industry are staggering and the personal costs have no price tag. Although the contributions from other <span id=\"urn:enhancement-4111bb35-a8ff-4df1-bbbd-a63012cc2988\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/the-nervous-system\">CNS<\/span> cell types, like reactive <span id=\"urn:enhancement-fab829c1-a498-439f-83b8-2d76c368532c\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/astrocytes\">astrocytes<\/span> and inflammatory <span id=\"urn:enhancement-905e6df2-69c2-49f3-b5cd-df60e1dc03da\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/microglia\">microglia<\/span>, are not to be discounted; recently published work by scientists in the UCSD <span id=\"urn:enhancement-d0aa8afd-e80b-4c9a-af68-9356b461f9f6\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/neuroscience\">Neuroscience<\/span> Department confirm a 20-year old observation at a molecular level, that dysfunctional <span id=\"urn:enhancement-1f2a93e5-f01a-44ef-b0cf-1e3a3c4dcece\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/brain-cells\">neuronal<\/span> trafficking is associated with the development of <span id=\"urn:enhancement-eb991e73-defb-41b9-a83b-430907c84394\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/alzheimers\">AD<\/span>.<\/p>\n<p><strong><a href=\"http:\/\/www.statista.com\/statistics\/452945\/mortality-rate-of-alzheimers-patients-in-the-us\/\" target=\"_blank\" rel=\"nofollow noopener\"><img decoding=\"async\" src=\"http:\/\/www.statista.com\/graphic\/1\/452945\/mortality-rate-of-alzheimers-patients-in-the-us.jpg\" alt=\"Statistic: Alzheimer's disease mortality rate in the United States from 2000 to 2013 (per 100,000 population) | Statista\" \/><\/a>Find more statistics at <a href=\"http:\/\/www.statista.com\/\" target=\"_blank\" rel=\"nofollow noopener\">Statista<\/a><\/strong><\/p>\n<p>The movement of materials from outside a cell or transport of synthesized proteins within a cell is generally termed \u201ccellular trafficking\u201d. Metaphorically, these processes represent a complex folk dance, with rapidly changing partners, rhythm and tempo changes and intricate steps. In <a href=\"https:\/\/www.mybiosource.com\/learn\/diseases\">diseases<\/a> like <span id=\"urn:enhancement-22ef23b2-53e5-4a72-83d6-7effa870f025\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/alzheimers\">AD<\/span>, these steps can be blocked, missing \u201cdance partners\u201d or have the music change direction. Cataldo et al., used microscopy to visualize one of these dance steps in <span id=\"urn:enhancement-5762f177-bb39-4a2b-9815-0ae7669e6914\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/pyramidal-neurons\">pyramidal neurons<\/span> in 1997 1. His group found that an early <span id=\"urn:enhancement-c2ef8764-be57-457f-854a-87eb9a27ddb1\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/vesicle\">vesicle<\/span> in cellular trafficking, the endosomal compartment, is enlarged in <span id=\"urn:enhancement-c7323b81-b921-484d-b8fa-ecbfe4eee6ea\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/alzheimers\">AD<\/span> neurons compared to non-disease controls. At the time, endocytotic machinery was just being elucidated and little was known about the functions and <span id=\"urn:enhancement-0d169076-1fdd-4515-8e52-3e514447f972\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/cell-signaling\">signaling<\/span> properties of these compartments. In subsequent years, we have gained a greater understanding of the processes from stimulation of membrane <span id=\"urn:enhancement-46efbf10-ad33-4aa8-9835-e0c986d94cf0\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/receptors\">receptors<\/span>, to early <span id=\"urn:enhancement-9d356fb7-f9be-4022-b04d-b0bc3cb09d54\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/intracellular\">intracellular<\/span> <span id=\"urn:enhancement-355c3ba4-7942-4300-af59-b029650d7dec\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/cell-signaling\">signaling<\/span> cascades, to the function of <span id=\"urn:enhancement-c05136d0-9cb0-405c-8b06-49ab6d33ef77\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/endosomes\">endosomes<\/span> as <span id=\"urn:enhancement-77bcaa91-6af9-4d33-8d2b-2fcae420f7ec\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/cell-signaling\">signaling<\/span> platforms, recycling compartments and sorting compartments.<\/p>\n<p>Many of these key trafficking steps have <span id=\"urn:enhancement-614e89a5-bbc1-4217-8f1c-8c172ac154e4\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/alzheimers\">AD<\/span> associated protein <span id=\"urn:enhancement-3c91d002-288e-4547-945a-10355f0a95ce\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/expression\">expression<\/span> changes or newly recognized <span id=\"urn:enhancement-4c293bbc-df14-464d-b669-2ac928091034\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/mutations\">mutations<\/span>. Early in the 1990\u2019s, the first major variant clearly associated with the development of <span id=\"urn:enhancement-3e7c0f32-a518-4b82-91c5-2cde8c73d029\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/alzheimers\">AD<\/span> was identified: <span id=\"urn:enhancement-bf7a9d83-50a8-4845-babd-659b23109fe7\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/apoe4\">ApoE4<\/span>. In the <span id=\"urn:enhancement-70c02167-093c-482a-9b23-a92220bab116\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/the-nervous-system\">CNS<\/span>, <span id=\"urn:enhancement-5fdd9a84-3cb5-438c-b5cf-64ee4cfa02b5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/apoe4\">ApoE<\/span> uptake is through <span id=\"urn:enhancement-566e5120-3358-4078-afd8-1e8422e4f3b7\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/clathrin\">clathrin<\/span> \u2013 mediated <span id=\"urn:enhancement-7bfe3103-58d0-4fc2-8bf9-5fec963935b1\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/endocytosis\">endocytosis<\/span> by <span id=\"urn:enhancement-b17300b1-7a53-436b-9f09-9c85498f5c1b\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/lrp1\">LRP1<\/span>, which has decreased <span id=\"urn:enhancement-45db734d-6486-4215-861c-7b3acc73681d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/expression\">expression<\/span> in <span id=\"urn:enhancement-2143b971-e94c-4ed8-a5e1-c31450cd6964\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/alzheimers\">AD<\/span> patients. The <span id=\"urn:enhancement-45da4067-a3bd-4dda-bec3-7369754de767\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/clathrin\">clathrin<\/span> coat is critical for later fusion with <span id=\"urn:enhancement-750b748e-425e-4ec1-bd5b-0dab32c1e164\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/endosomes\">endosomes<\/span>. PICALM, with known highly penetrant <span id=\"urn:enhancement-2057f194-31b2-4de6-b3eb-9864e2721043\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/alzheimers\">AD<\/span> associated <span id=\"urn:enhancement-e454570e-b138-421d-a294-32f2e0691282\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/mutations\">mutations<\/span>, recruits <span id=\"urn:enhancement-693892f8-da1d-460f-a025-230627ca9d10\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/clathrin\">clathrin<\/span> to the membrane. <span id=\"urn:enhancement-44fd8fdc-4682-4005-898c-d8177f438699\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/apoe4\">ApoE<\/span> further traffics to early <span id=\"urn:enhancement-b8aeddcb-8419-44d9-8752-da4849e8a3c8\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/endosomes\">endosomes<\/span> and can take one of several subsequent routes through late <span id=\"urn:enhancement-1ceb3fee-dcba-4dfd-8f0f-a9f6c7dc6a03\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/endosomes\">endosomes<\/span> to golgi stacks or directly to lysosomes for degradation. The current study focuses in on early <span id=\"urn:enhancement-412d68eb-f02b-4d12-a3fe-9cbba3994e8e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/endosomes\">endosomes<\/span>, marked by the small molecular switch Rab5. When Rab5 is constitutively \u201con\u201d, <span id=\"urn:enhancement-a1da45f4-c9bf-4637-ae76-e896a831b311\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/endosomes\">endosomes<\/span> become enlarged since cargo isn\u2019t handed off in a precisely timed fashion. The researchers at UCSD have found that Rab5 is abnormally activated in <span id=\"urn:enhancement-2861f348-3704-4d6c-953f-88e0eb2c7e11\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/alzheimers\">AD<\/span>, causing early <span id=\"urn:enhancement-ad13b650-a6b5-4276-9cfd-4b41aa7758e6\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/cell-signaling\">signaling<\/span> and trafficking dysfunction, just one more piece in the trafficking puzzle 2. With cargo movement being blocked or hindered in this early transition, neurons are deprived of nuclear <span id=\"urn:enhancement-f33ed31e-0329-4b6f-81c7-9f357f34d654\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/translocation\">translocation<\/span> for <span id=\"urn:enhancement-95a2793d-82ad-4e69-98e3-17c52a72133a\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/transcriptional-regulation\">transcriptional regulation<\/span>, cargo transport of cellular nutrients and required recycling or degradation of membrane <span id=\"urn:enhancement-f35099ed-32d7-4322-8dc3-78bafd04afb5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/receptors\">receptors<\/span>.<\/p>\n<p>1.
