The most unlikely of findings can propel fields to entirely unexpected directions. A recent example is highlighted by work from a group of cell biologists at UC-San Diego. In the on-line journal eLife, these researchers connect a well-characterized <span id=\"urn:enhancement-b87afa2d-d9aa-4fcc-bd38-3a5fef729678\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/metabolic-pathways\">metabolic pathway<\/span>, LKB1-<span id=\"urn:enhancement-beb3fd70-53e2-4753-8b24-9f4f8ad04df4\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span>, to a previously unknown effector protein, GIV and in doing so uncover very significant discoveries about <span id=\"urn:enhancement-880fea58-ca6c-444c-aac0-db58d366c430\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/invasive\">cancer<\/span> biology. First they reveal a long sought after anti-<span id=\"urn:enhancement-403a526b-2aa6-4e18-87af-ac46ea19c19a\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/invasive\">cancer<\/span> mechanism for the widely used diabetes drug, <span id=\"urn:enhancement-8a93eb0e-4a64-44d9-adf3-860b604479cc\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/metformin\">Metformin<\/span> and next they link tumor development and metastases to possible <span id=\"urn:enhancement-ec033cfc-dd65-4f11-9e7f-53c16e232226\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/mutations\">mutations<\/span> in this novel pathway.<img loading=\"lazy\" decoding=\"async\" class=\"alignright size-medium wp-image-321\" src=\"https:\/\/www.mybiosource.com\/learn\/wp-content\/uploads\/2017\/03\/cancer-biology-300x213.jpg\" alt=\"\" width=\"300\" height=\"213\" srcset=\"https:\/\/www.mybiosource.com\/learn\/wp-content\/uploads\/2017\/03\/cancer-biology-300x213.jpg 300w, https:\/\/www.mybiosource.com\/learn\/wp-content\/uploads\/2017\/03\/cancer-biology-370x263.jpg 370w, https:\/\/www.mybiosource.com\/learn\/wp-content\/uploads\/2017\/03\/cancer-biology.jpg 450w\" sizes=\"(max-width: 300px) 100vw, 300px\" \/><\/p>\n<h3>Fences keep neighborhoods friendly<\/h3>\n<p>We build fences to keep things out and our bodies have a similar approach. Specialized epithelial cells form tight and rigid barriers to protect areas such as the gut, lungs, brain and oral cavities. Their structure is imposed by rigid tight junction (TJ) complexes, which connect neighboring cells together. This organization also imposes restricted movement and restricted <span id=\"urn:enhancement-7dd26241-8c8a-4064-9e95-cbb9d93676be\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/cell-proliferation\">cell proliferation<\/span>. As single cell layers, each surface is exposed to a different environment, creating an asymmetrical polarized cell. The apical end is outward facing and the basal surface attaches to the tissue below. Apical and basal surfaces perform critical and distinct cellular functions and when this polarity is lost, the barrier falls apart.<\/p>\n<h3>How these pathways collide: AMPK is the hammer and GIV is the nail<\/h3>\n<p>Our bodies run on energy derived from <span id=\"urn:enhancement-010fb1ca-26bc-488e-89f1-422325dacaad\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/atp\">ATP<\/span>. Degradation of <span id=\"urn:enhancement-d4dc44cc-f0b3-4e9d-ad6e-e869a0b521fd\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/atp\">ATP<\/span> creates the <span id=\"urn:enhancement-fd0d6a55-f7c7-47cb-adc4-64e51499ad3a\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/intracellular\">intracellular<\/span> byproduct <span id=\"urn:enhancement-bedcf609-3c7c-4def-9422-00648b44a046\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/amp\">AMP<\/span>. The ratios of <span id=\"urn:enhancement-23301d81-7fc4-4294-aa9e-3216725d7324\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/atp\">ATP<\/span> to <span id=\"urn:enhancement-848ad1f5-5d02-422b-879e-9ca9dac50eec\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/amp\">AMP<\/span> relate to the energy state of individual cells in time. <span id=\"urn:enhancement-178676e6-bc33-439d-b810-457413878917\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span>, <span id=\"urn:enhancement-51daf5e8-49bc-455c-ba78-2b4d6cdffb8e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/amp\">AMP<\/span>-activated <span id=\"urn:enhancement-227e414b-be55-48be-b52c-a49d54f8b40f\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/protein-kinase\">protein kinase<\/span>, is the critical sensor of this ratio and is phosphorylated by LKB1 during states of energetic stress when <span id=\"urn:enhancement-a8325c9e-a28a-4a15-be5d-9909eae6b2c9\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/amp\">AMP<\/span> concentration rises.<\/p>\n<p>In polarized epithelial cells, maintenance of the structural TJ requires significant energy costs, making regulation of the LKB1-<span id=\"urn:enhancement-28dc08e6-592f-4134-99df-ddeffeef7ea7\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span> pathway critical for homeostasis. Either functional loss or <span id=\"urn:enhancement-a7e8aa4e-f738-4c7a-a602-aab3a9831542\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/expression\">expression<\/span> reductions in <span id=\"urn:enhancement-93779470-3aa0-4318-905d-63011666e293\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span> are associated with a breakdown in epithelial linings by a weakening of the TJ complex. Although this relationship has been well described, the precise molecular mechanism for <span id=\"urn:enhancement-2afeea85-8c36-407a-8c9e-06bd6e663c8a\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span> protection of TJ complexes was still mysterious.<\/p>\n<p>Sitting at the intersection between <span id=\"urn:enhancement-18a46dc5-2b05-4677-b8a6-71d996a8e9c0\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/intracellular\">intracellular<\/span> <span id=\"urn:enhancement-22559eef-1a7d-4484-b216-4cf5bcbc692e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span> and the membrane localized TJ, is the multimodular scaffolding protein GIV\/Girdin. Originally discovered in 2005, GIV (<span id=\"urn:enhancement-c24c3bd4-0760-42cd-8584-6f57fb9d4b1f\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/g-alpha\">G-alpha<\/span> interacting vesicle associated protein) is a large and complex enigmatic protein. Other multifunctional proteins are known, but GIV takes multi-tasking to the extreme. The C-terminus has both <span id=\"urn:enhancement-314eb05f-9a4b-42a2-8121-a979d7771b39\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/gef\">GEF<\/span> (<span id=\"urn:enhancement-d2b37410-978c-4b4b-8d3b-ef5ef25f6d37\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/guanine\">guanine<\/span> nucleotide exchange factor) and GDI (<span id=\"urn:enhancement-500e3f5e-7de0-4e7c-99ac-4f30d1b5b83d\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/guanine\">guanine<\/span> nucleotide dissociation inhibitor) capabilities to regulate <span id=\"urn:enhancement-d7ea89fc-7234-4be7-8666-bdba741854fc\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/g-protein\">G-protein<\/span> signaling. While the <span id=\"urn:enhancement-00ac9574-c6b1-40e2-a00e-65e628bab0c8\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/n-terminal\">N-terminus<\/span> has some TJ protein binding domains, no one had put <span id=\"urn:enhancement-00c737da-3ab7-4e47-8417-fc42a1a53412\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span> and GIV together in a single pathway until now.<\/p>\n<p>Using bioinformatics to determine putative phosphorylation sites within the <span id=\"urn:enhancement-e8ac1035-5288-4162-ab6e-8cdfb474c096\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/n-terminal\">N-terminus<\/span>, these scientists, lead by senior author Dr. Pradipta Ghosh, MD, found one specific <span id=\"urn:enhancement-2b821a58-e587-4e6a-8442-b8e928e47b73\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/serine\">serine<\/span> (S245) was directly phosphorylated by activated <span id=\"urn:enhancement-ce45a88f-c2d2-408c-9f49-119c322f1094\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span> in response to energetic stress in cultured epithelial cells. <span id=\"urn:enhancement-2f830929-b489-47a8-bd28-c6de70f95c94\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/phosphorylation\">Phosphorylation<\/span> of S245 on GIV was sufficient to stabilize TJ complexes and maintain barrier functions, a key missing link in the known mechanism of <span id=\"urn:enhancement-048da424-ed1c-4eb2-b45c-2d459979a8d0\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span>-mediated protection.<\/p>\n<h3>Why stability matters<\/h3>\n<p>With a mechanism for TJ stability in hand, Ghosh et al, tackled a long standing mystifying observation of clinicians: long-term treatment with the <span id=\"urn:enhancement-7005b3c2-b1c8-4187-b20f-b85e883a6543\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span> activator <span id=\"urn:enhancement-020f353b-f924-4cb0-8b6f-0b25c3679460\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/metformin\">Metformin<\/span> (Glucophage) has beneficial anti-<span id=\"urn:enhancement-4c9fe645-af86-4671-b14d-f25fbc8a7957\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/invasive\">cancer<\/span> side effects. Using <span id=\"urn:enhancement-4aff6387-ceae-44f7-b5e2-5b5fd05335cf\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/metformin\">Metformin<\/span> as a direct activator for <span id=\"urn:enhancement-47220351-599d-44a2-b689-0ae022cbe68e\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span> or Compound C, a known inhibitor of <span id=\"urn:enhancement-69907152-ace0-41f0-9031-bdafbccc1f77\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span> activation as a negative control, they were able to demonstrate that specific phosphorylation of GIV S245 increases stability of TJs even in glucose starvation <a href=\"https:\/\/www.mybiosource.com\/learn\/conditions\/\">conditions<\/a>. Stability equates to homeostasis. Initiation of oncogenic progression is preceded by a loss of cellular polarity and <span id=\"urn:enhancement-4c299204-6467-45cf-834c-fced26e076f5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/metformin\">Metformin<\/span> apparently staves off this cellular change by activating the <span id=\"urn:enhancement-5d3c259a-a6c6-4b78-a5ed-c085a8ea0058\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span>-GIV pathway.<\/p>\n<p>However, while this tumor suppression finding is optimistic and encouraging, there is a dark side as well. While screening several <span id=\"urn:enhancement-f13e6b71-5906-4736-b174-6561b5c696f5\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/invasive\">cancer<\/span> databases for GIV <span id=\"urn:enhancement-153a6c06-c44a-4347-a1fc-64be2313ec72\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/mutations\">mutations<\/span>, areas within and surrounding the <span id=\"urn:enhancement-ee90554b-bb20-4bfe-9354-27eaed1ee978\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span>-binding domain frequently popped up with noteworthy changes. One particular mutation, L249P, four residues from S245 and within the consensus-<span id=\"urn:enhancement-1839108c-1cdb-4959-8fc0-f599cde818ce\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/binding-sites\">binding site<\/span> for <span id=\"urn:enhancement-f115b366-48d0-465e-840d-e1e28277e265\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/ampk\">AMPK<\/span>, abolishes the phosphorylation of GIV by allosteric hindrance. Cell expressing this mutant GIV will fail to stabilize TJs under energetic stress, will lose polarity and can initiate early oncogenic changes.
