MBS2023550 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Leukocyte Elastase Inhibitor (LEI) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing LEI. The ELISA analytical biochemical technique of the MBS2023550 kit is based on LEI antibody-LEI antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect LEI antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, LEI. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Leukocyte Elastase Inhibitor (LEI). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Leukocyte Elastase Inhibitor (LEI). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Leukocyte Elastase Inhibitor (LEI), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Leukocyte Elastase Inhibitor (LEI) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene is a member of the serpin family of proteinase inhibitors. Members of this family maintain homeostasis by neutralizing overexpressed proteinase activity through their function as suicide substrates. This protein inhibits the neutrophil-derived proteinases neutrophil elastase, cathepsin G, and proteinase-3 and thus protects tissues from damage at inflammatory sites.
