MBS564009 is a ready-to-use microwell, strip-or-full plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Chicken Ovotransferrin (LTF) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing LTF. The ELISA analytical biochemical technique of the MBS564009 kit is based on LTF antibody-LTF antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect LTF antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, LTF. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%). This product is a highly sensitive ELISA based assay kit for detecting and quantifying chicken ovotransferrin (conalbumin). In this indirect ovotransferrin conalbumin ELISA assay, known amounts of anti-chicken ovotransferrin antibodies are adsorbed to each plate well. A sample containing chicken ovotransferrin is added to each well and binds to capture antibodies. Unbound antigen is washed away. Subsequently, a detection antibody solution containing anti-ovotransferrin antibodies conjugated with horseradish peroxidase is added, forming a complex with the previously bound chicken ovotransferrin and anti-ovotransferrin capture antibodies. Another wash removes unbound detection antibodies, and a chromogenic substrate is added. The amount of bound enzyme varies directly with the concentration of protein in the original test sample, thus the absorbance at 450nm is inversely proportional with the concentration of conalbumin added to each well. The concentration of ovotransferrin in the test sample can be derived from a calibration curve of known standards.

Intended Uses: The total Chicken Ovotransferrin (TX) test kits are a highly sensitive two-site enzyme linked immunoassay (ELISA) for measuring Ovotransferrin in biological samples of Chickens.

Principle of the Assay: The principle of the double antibody sandwich ELISA is represented in Figure 1. In this assay the TX present in samples reacts with the anti-TX antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-TX antibodies conjugated with horseradish peroxidase (HRP) are added. These enzyme-labeled antibodies form complexes with the previously bound TX. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of TX in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of TX in the test sample. The quantity of TX in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.
