Species Reactivity
Specificity
This assay has high sensitivity and excellent specificity for detection of MMP-12. No significant cross-reactivity or interference between MMP-12 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between MMP-12 and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
1.0 pg/mL.
Preparation and Storage
Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note
Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of MMP-12 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS750829 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Matrix metalloproteinase 12 (MMP-12) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing MMP-12. The ELISA analytical biochemical technique of the MBS750829 kit is based on MMP-12 antibody-MMP-12 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MMP-12 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MMP-12. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This MMP-12 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine MMP-12. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: MMP-12 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-MMP-12 antibody and an MMP-12-HRP conjugate. The assay sample and buffer are incubated together with MMP-12-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MMP-12 concentration since MMP-12 from samples and MMP-12-HRP conjugate compete for the anti-MMP-12 antibody binding site. Since the number of sites is limited, as more sites are occupied by MMP-12 from the sample, fewer sites are left to bind MMP-12-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MMP-12 concentration in each sample is interpolated from this standard curve.

NCBI Summary for MMP-12
Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. It is thought that the protein encoded by this gene is cleaved at both ends to yield the active enzyme, but this processing has not been fully described. The enzyme degrades soluble and insoluble elastin. It may play a role in aneurysm formation and studies in mice suggest a role in the development of emphysema. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3. [provided by RefSeq, Jul 2008]
UniProt Comments for MMP-12
MMP12: May be involved in tissue injury and remodeling. Has significant elastolytic activity. Can accept large and small amino acids at the P1' site, but has a preference for leucine. Aromatic or hydrophobic residues are preferred at the P1 site, with small hydrophobic residues (preferably alanine) occupying P3. Belongs to the peptidase M10A family.
