The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing MMP-8. The ELISA analytical biochemical technique of the MBS726999 kit is based on MMP-8 antibody-MMP-8 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MMP-8 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MMP-8. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This MMP-8 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse MMP-8. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: MMP-8 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-MMP-8 antibody and an MMP-8-HRP conjugate. The assay sample and buffer are incubated together with MMP-8-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MMP-8 concentration since MMP-8 from samples and MMP-8-HRP conjugate compete for the anti-MMP-8 antibody binding site. Since the number of sites is limited, as more sites are occupied by MMP-8 from the sample, fewer sites are left to bind MMP-8-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MMP-8 concentration in each sample is interpolated from this standard curve.

Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. However, the enzyme encoded by this gene is stored in secondary granules within neutrophils and is activated by autolytic cleavage. Its function is degradation of type I, II and III collagens. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3. [provided by RefSeq, Jul 2008]
