MBS703279 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the matrix metallopeptidase 1 (interstitial collagenase) (MMP1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing MMP1. The ELISA analytical biochemical technique of the MBS703279 kit is based on MMP1 antibody-MMP1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MMP1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MMP1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MMP-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MMP-1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP-1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP-1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. This gene encodes a secreted enzyme which breaks down the interstitial collagens, types I, II, and III. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3. Alternative splicing results in multiple transcript variants.[provided by RefSeq, Mar 2009]

Cofactor: Binds 4 calcium ions per subunit.
