MBS824765 is a ready-to-use microwell, strip-or-full plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the MMP12, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing MMP12. The ELISA analytical biochemical technique of the MBS824765 kit is based on MMP12 antibody-MMP12 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MMP12 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MMP12. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: Mouse MMP12 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse MMP12 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse MMP12 coated on a 96-well plate. Standards and samples are pipetted into the wells and MMP12 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse MMP12 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MMP12 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Matrix metalloproteinase-12 (MMP12), also known as MME or ME, is an enzyme that in humans is encoded by the MMP12 gene. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.2. It is thought that the protein encoded by this gene is cleaved at both ends to yield the active enzyme, but this processing has not been fully described. The enzyme degrades soluble and insoluble elastin. It may play a role in aneurysm formation and studies in mice suggest a role in the development of emphysema. This gene may involved in tissue injury and remodeling.

UniProt Comments for MMP12
MMP12: May be involved in tissue injury and remodeling. Has significant elastolytic activity. Can accept large and small amino acids at the P1' site, but has a preference for leucine. Aromatic or hydrophobic residues are preferred at the P1 site, with small hydrophobic residues (preferably alanine) occupying P3. Belongs to the peptidase M10A family.
