Product Description: The monoclonal antibody 16D2 recognizes mouse complement factor C4, formerly known as Gg protein, which consists of an alpha-, beta-, and gamma-chain. The classical pathway of complement and the Mannose binding lectin activation pathway converge at C4. C1s, MASP-1 and MASP-2 cleave C4 resulting in the formation of C4a and C4b. Subsequently, C4b can be cleaved to C4c and C4d by other serum enzymes. The monoclonal antibody 16D2 reacts with intact C4, C4b and C4d.
C4 is an acute phase protein that is produced by hepatocytes, monocytes and intestinal epithelial cells and can be used- in experimental animals as a marker for activation of the classical complement pathway. Recent studies have demonstrated an association between graft rejection and C4d deposition in a mouse model for cardiac transplantation.

Application Notes for anti-C4 antibody
Application Notes: W: A non-reduced sample treatment and SDS-Page was used. The band size is 200x kDa (Ref.4).
IA: Plates were coated overnight with rat anti-murine C4 mAb 16D2 in carbonate buffer and blocked with 5% dry milk in PBS and 0.01% Tween 20.
IF: Spleens were snap frozen and stored at -80 degree C until cryosections were cute. (Section (5um) were fixed for 4 mins in ice-cold acetone. Sections were blocked with 2% BSA, 2% FCS and PBS. Biotinylated 16D2 was visualized using avidin-PE.
Application Use: For immunohistochemistry and Western blotting, dilutions to be used depend on detection system applied. It is recommended that users test the reagent and determine their own optimal dilutions. The typical starting working dilution is 1:50.

C4B: C4 plays a central role in the activation of the classical pathway of the complement system. It is processed by activated C1 which removes from the alpha chain the C4a anaphylatoxin. The remaining alpha chain fragment C4b is the major activation product and is an essential subunit of the C3 convertase (C4b2a) and the C5 convertase (C3bC4b2a) enzymes of the classical complement pathway. Defects in C4B are a cause of susceptibility to systemic lupus erythematosus (SLE). A chronic, inflammatory and often febrile multisystemic disorder of connective tissue. It affects principally the skin, joints, kidneys and serosal membranes. It is thought to represent a failure of the regulatory mechanisms of the autoimmune system. Interindividual copy- number variation (CNV) of complement component C4 and associated polymorphisms result in different susceptibilities to SLE. The risk of SLE susceptibility has been shown to be significantly increased among subjects with only two copies of total C4. A high copy number is a protective factor against SLE. Defects in C4B are the cause of complement component 4B deficiency (C4BD). A rare defect of the complement classical pathway associated with the development of autoimmune disorders, mainly systemic lupus with or without associated glomerulonephritis.
