Store at 4 degree C or at -20 degree C if preferred. This product should be stored undiluted. Storage in frost-free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

Small volumes of anti-GZMB antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Mouse anti Granzyme B antibody, clone GB11 recognizes the serine protease Granzyme B, which is important in the induction of apoptosis in target cells by cytolytic lymphocytes (CTLs). Granzyme B plays a key role in the induction of apoptosis by CTLs. After delivery to the target cell, Granzyme B activates the cascade of caspases that finally results in cell death. Mouse anti Granzyme B antibody, clone GB11 is suitable for the detection of Granzyme B expressing cells by flow cytometry. In normal peripheral blood approximately 20% of CD8+ve T cells have been found to express Granzyme B.

Application Notes for anti-GZMB antibody
ELISA: This antibody has been reported to function as a capture reagent in sandwich ELISA assays for soluble Granzyme B in conjunction with biotin conjugated clone GB10 as detection reagent.
Flow Cytometry: Use 10ul of the suggested working dilution to label 106 cells or 100ul whole blood.
ELISA: Minimum Dilution: 2ug/ml; Maximum Dilution: 5ug/ml
Flow Cytometry: Minimum Dilution: 1/10; Maximum Dilution: 1/100; Application Note: Membrane permeabilisation is required for this application. MyBioSource recommends the use of Leucoperm for this purpose.

Cytolytic T lymphocytes (CTL) and natural killer (NK) cells share the remarkable ability to recognize, bind, and lyse specific target cells. They are thought to protect their host by lysing cells bearing on their surface 'nonself' antigens, usually peptides or proteins resulting from infection by intracellular pathogens. The protein encoded by this gene is crucial for the rapid induction of target cell apoptosis by CTL in cell-mediated immune response. [provided by RefSeq, Jul 2008]
