Recognizes human alpha-skeletal and alpha-cardiac (sarcomeric) actins as well as neoplasms derived from normal tissues (i.e. rhabdomyosarcomas). Shows no crossreaction with smooth muscle actin. Species Crossreactivity: bovine, rabbit, sheep, rat, guinea pig, and frog.
Purified by ammonium sulfate precipitation.
Supplied as a liquid in PBS, pH 7.4. Also available with BSA and azide. See A0760-24.
N-terminal peptide of alpha skeletal muscle actin; acetylated at the N-terminus and conjugated to KLH at the C-terminus. Cellular Localization: Cytoplasmic. Epitope: N-terminal decapeptide.

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Other Notes
Small volumes of anti-ACTA1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

Suitable for use in Immunofluorescence and Immunohistochemistry.
Dilution: Immunohistochemistry: 2-4ug/ml for 30 min. at RT.
Formalin/paraffin: No special pretreatment is required for the immunohistochemical staining of formalin/paraffin tissues.

Function: Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells.

Subunit structure: Polymerization of globular actin (G-actin) leads to a structural filament (F-actin) in the form of a two-stranded helix. Each actin can bind to 4 others.

Subcellular location: Cytoplasm › cytoskeleton.

Post-translational modification: Oxidation of Met-46 and Met-49 by MICALs (MICAL1, MICAL2 or MICAL3) to form methionine sulfoxide promotes actin filament depolymerization. MICAL1 and MICAL2 produce the (R)-S-oxide form. The (R)-S-oxide form is reverted by MSRB1 and MSRB2, which promote actin repolymerization

Miscellaneous: In vertebrates 3 main groups of actin isoforms, alpha, beta and gamma have been identified. The alpha actins are found in muscle tissues and are a major constituent of the contractile apparatus. The beta and gamma actins coexist in most cell types as components of the cytoskeleton and as mediators of internal cell motility.
