Performance
Five x 105 cultured HPB-MLT human tumor cells were washed and incubated 45 minutes on ice with 80 mul of anti-CD134/R-PE at a 1:50 dilution (5 mug/ml). Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 0.85 log10 fluorescent units when compared to a Mouse IgG1/R-PE negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20 mul of 0.5 mg/ml anti-CD134 antibody.

Protein A purified antibody from tissue culture supernatant was conjugated to R-Phycoerythrin through a sulfo-ester linkage. Unconjugated antibody was removed using size exclusion chromatography. Antibody conjugate is at 0.25 mg/ml with a A565/A280 ratio of 3.0.

Store at 2 - 5 degree C. Do not freeze! Protect from light.

If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

Information: Human CD134 (OX40) is an activation-associated antigen which is predominantly expressed on activated CD4 positive cells. CD134 antigen is a member of the tumor necrosis factor (TNF) receptor family of molecules and may be involved with regulating T cell-dependent B cell proliferation and differentiation. Antibody Ber Act 35 recognizes the human CD134 molecule of about 35 kd.

The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor has been shown to activate NF-kappaB through its interaction with adaptor proteins TRAF2 and TRAF5. Knockout studies in mice suggested that this receptor promotes the expression of apoptosis inhibitors BCL2 and BCL2lL1/BCL2-XL, and thus suppresses apoptosis. The knockout studies also suggested the roles of this receptor in CD4+ T cell response, as well as in T cell-dependent B cell proliferation and differentiation. [provided by RefSeq, Jul 2008]
