anti-rat CD8a monoclonal antibody recognizes a determinant on the majority of thymocytes (90-95%), a subset of peripheral T cells, the majority of NK cells, and the granular intraepithelial leukocytes in the small intestine. (1,2,3,4.) The antigen recognized is a complex of surface glycoproteins of Mr 34,000, 39,000 and 76,000 Da and is the rat homologue of the human CD8 and the mouse Ly-2,3 antigen. (5,8) MBS520223 labels all peripheral T cells that are unlabelled by the (W3/25) monoclonal antibody. It labels a T-cell subset which mediates the suppression of antibody formation (1) and the cytotoxic cell precursor. (2) MBS520223 can be used together to fractionate T cells by sorting in FACS or by rosette depletion (1,2) or can be used together to study subsets of T cells in the rat which mediate lethal graft vs. host disease. (7) This antibody is one of the 3 monoclonal antibodies which labels T lymphocyte populations in the rat, these being (W3/13) which labels all T cells, as well as (W3/25) and (MRC 0X-8) which label non-overlapping T cell populations. These monoclonal antibodies used in concert are being employed extensively to investigate cellular aspects of the immune response in rats and prove to be useful as markers for functionally distinct subpopulations of lymphocytes. This clone is reported to work with frozen and paraffin sections (9).

The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. The CD8 antigen acts as a coreceptor with the T-cell receptor on the T lymphocyte to recognize antigens displayed by an antigen presenting cell in the context of class I MHC molecules. The coreceptor functions as either a homodimer composed of two alpha chains or as a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. This gene encodes the CD8 alpha chain. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Nov 2011]

Shown to be expressed as heterodimer on thymocytes and as homodimer on peripheral blood T-lymphocytes. Interacts with the MHC class I HLA-A/B2M dimer. Interacts with LCK in a zinc-dependent manner.
