Species Reactivity
Specificity
This assay has high sensitivity and excellent specificity for detection of MPIF1. No significant cross-reactivity or interference between MPIF1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between MPIF1 and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage
Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note
Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of MPIF-1 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS721461 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Myeloid Progenitor Inhibitory Factor 1 (MPIF-1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing MPIF-1. The ELISA analytical biochemical technique of the MBS721461 kit is based on MPIF-1 antibody-MPIF-1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MPIF-1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MPIF-1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This MPIF1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse MPIF1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: MPIF1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MPIF1 antibody and an MPIF1-HRP conjugate. The assay sample and buffer are incubated together with MPIF1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MPIF1 concentration since MPIF1 from samples and MPIF1-HRP conjugate compete for the anti-MPIF1 antibody binding site. Since the number of sites is limited, as more sites are occupied by MPIF1 from the sample, fewer sites are left to bind MPIF1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MPIF1 concentration in each sample is interpolated from this standard curve.

NCBI Summary for MPIF-1
This gene is one of several chemokine genes clustered on the q-arm of chromosome 17. Chemokines form a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of the N-terminal cysteine residues of the mature peptide. This chemokine, a member of the CC subfamily, displays chemotactic activity on resting T lymphocytes and monocytes, lower activity on neutrophils and no activity on activated T lymphocytes. The protein is also a strong suppressor of colony formation by a multipotential hematopoietic progenitor cell line. In addition, the product of this gene is a potent agonist of the chemokine (C-C motif) receptor 1. Alternative splicing results in multiple transcript variants that encode different isoforms. UniProt Comments for MPIF-1
CCL6: Shows chemotactic activity for monocytes, resting T- lymphocytes, and neutrophils, but not for activated lymphocytes. Inhibits proliferation of myeloid progenitor cells in colony formation assays. This protein can bind heparin. Binds CCR1. CCL23(19-99), CCL23(22-99), CCL23(27-99), CCL23(30-99) are more potent chemoattractants than the small-inducible cytokine A23. Belongs to the intercrine beta (chemokine CC) family. 2 isoforms of the human protein are produced by alternative splicing.
