Species Reactivity
Specificity
This assay has high sensitivity and excellent specificity for detection of human MUC3A. No significant cross-reactivity or interference between human MUC3A and analogues was observed.
Samples
Serum, plasma, tissue homogenates
Assay Type
Sandwich
Detection Range
1.56 ng/ml -100 ng/ml.
Sensitivity
The minimum detectable dose of human MUC3A is typically less than 0.39 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision
Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision
Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
Preparation and Storage
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
ISO Certification
Manufactured in an ISO 9001:2008 Certified Laboratory.
Product Note
Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of MUC3A elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS930725 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the mucin 3A, cell surface associated (MUC3A) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing MUC3A. The ELISA analytical biochemical technique of the MBS930725 kit is based on MUC3A antibody-MUC3A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MUC3A antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MUC3A. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MUC3A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MUC3A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MUC3A is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MUC3A bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Summary for MUC3A
The mucin genes encode epithelial glycoproteins, some of which are secreted and some membrane bound. Each of the genes contains at least one large domain of tandemly repeated sequence that encodes the peptide sequence rich in serine and/or threonine residues, which carries most of the O-linked glycosylation (Gendler and Spicer, 1995 [PubMed 7778880]).[supplied by OMIM, Aug 2008]
UniProt Comments for MUC3A
MUC3A: Major glycoprotein component of a variety of mucus gels. Thought to provide a protective, lubricating barrier against particles and infectious agents at mucosal surfaces. May be involved in ligand binding and intracellular signaling. 5 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, integral

Chromosomal Location of Human Ortholog: 7q22

Cellular Component: Golgi lumen; extracellular region; integral to membrane

Molecular Function: extracellular matrix structural constituent; extracellular matrix constituent, lubricant activity

Biological Process: protein amino acid O-linked glycosylation; cellular protein metabolic process; O-glycan processing; post-translational protein modification
