The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing NPHN. The ELISA analytical biochemical technique of the MBS2022646 kit is based on NPHN antibody-NPHN antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect NPHN antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, NPHN. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Nephrin (NPHN). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Nephrin (NPHN). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Nephrin (NPHN), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Nephrin (NPHN) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a member of the immunoglobulin family of cell adhesion molecules that functions in the glomerular filtration barrier in the kidney. The gene is primarily expressed in renal tissues, and the protein is a type-1 transmembrane protein found at the slit diaphragm of glomerular podocytes. The slit diaphragm is thought to function as an ultrafilter to exclude albumin and other plasma macromolecules in the formation of urine. Mutations in this gene result in Finnish-type congenital nephrosis 1, characterized by severe proteinuria and loss of the slit diaphragm and foot processes.[provided by RefSeq, Oct 2009]
