MBS701505 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the nuclear receptor subfamily 1, group H, member 2 (NR1H2) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing NR1H2. The ELISA analytical biochemical technique of the MBS701505 kit is based on NR1H2 antibody-NR1H2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect NR1H2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, NR1H2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for LXRbeta has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any LXRbeta present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LXRbeta is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LXRbeta bound in the initial step. The color development is stopped and the intensity of the color is measured.

The liver X receptors, LXRA (NR1H3; MIM 602423) and LXRB, form a subfamily of the nuclear receptor superfamily and are key regulators of macrophage function, controlling transcriptional programs involved in lipid homeostasis and inflammation. The inducible LXRA is highly expressed in liver, adrenal gland, intestine, adipose tissue, macrophages, lung, and kidney, whereas LXRB is ubiquitously expressed. Ligand-activated LXRs form obligate heterodimers with retinoid X receptors (RXRs; see MIM 180245) and regulate expression of target genes containing LXR response elements (summary by Korf et al., 2009 [PubMed 19436111]).[supplied by OMIM, Jan 2010]
