The ELISA Kit is designed to detect native, not recombinant, NR1H4. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. This assay employs a two-site sandwich ELISA to quantitate NR1H4 in samples. An antibody specific for NR1H4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NR1H4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NR1H4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NR1H4 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Ligand-activated transcription factor. Receptor for bile acids (BAs) such as chenodeoxycholic acid (CDCA), lithocholic acid, deoxycholic acid (DCA) and allocholic acid (ACA). Plays a essential role in BA homeostasis through the regulation of genes involved in BA synthesis, conjugation and enterohepatic circulation. Also regulates lipid and glucose homeostasis and is involved innate immune response. The FXR-RXR heterodimer binds predominantly to farnesoid X receptor response elements (FXREs) containing two inverted repeats of the consensus sequence 5'-AGGTCA-3' in which the monomers are spaced by 1 nucleotide (IR-1) but also to tandem repeat DR1 sites with lower affinity, and can be activated by either FXR or RXR-specific ligands. It is proposed that monomeric nuclear receptors such as NR5A2/LRH-1 bound to coregulatory nuclear responsive element (NRE) halfsites located in close proximity to FXREs modulate transcriptional activity.
