MBS2502608 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the PAR3, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PAR3. The ELISA analytical biochemical technique of the MBS2502608 kit is based on PAR3 antibody-PAR3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PAR3 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PAR3. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Chicken PAR3 concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to PAR3. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for PAR3 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain PAR3, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of PAR3. You can calculate the concentration of PAR3 in the samples by comparing the OD of the samples to the standard curve.

Polarity complex proteins, including PARD3, are essential for PIP3-induced receptor tyrosine kinase recycling. Seems to play a central role in the formation of epithelial tight junctions. Targets the phosphatase PTEN to cell junctions. Association with PARD6B may prevent the interaction of PARD3 with JAM1, thereby preventing tight junction assembly. The PARD6-PARD3 complex links GTP-bound Rho small GTPases to atypical protein kinase C proteins. Required for establishment of neuronal polarity and normal axon formation in cultured hippocampal neurons. PDZ 1 domain modulates interactions with JAM1, PARD6A and PARD6B; PDZ 2 domain mediates interaction with membranes containing phosphoinositol lipids; PDZ 3 domain regulates interactions with PTEN (via C-terminus). Belongs to the PAR3 family. 10 isoforms of the human protein are produced by alternative splicing. Isoform 2, but not at least isoform 3 interacts with PRKCZ.
