The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PCSK2. The ELISA analytical biochemical technique of the MBS2000087 kit is based on PCSK2 antibody-PCSK2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PCSK2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PCSK2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Proprotein Convertase Subtilisin/Kexin Type 2 (PCSK2). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Proprotein Convertase Subtilisin/Kexin Type 2 (PCSK2). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Proprotein Convertase Subtilisin/Kexin Type 2 (PCSK2), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Proprotein Convertase Subtilisin/Kexin Type 2 (PCSK2) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a member of the subtilisin-like proprotein convertase family, which includes proteases that process protein and peptide precursors trafficking through regulated or constitutive branches of the secretory pathway. The protein undergoes an initial autocatalytic processing event and interacts with a neuroendocrine secretory protein in the ER, exits the ER and sorts to secretory granules, where it is cleaved and catalytically activated during intracellular transport. The encoded protease is packaged into and activated in dense core secretory granules and expressed in the neuroendocrine system and brain. This gene encodes one of the seven basic amino acid-specific members which cleave their substrates at single or paired basic residues. It functions in the proteolytic activation of polypeptide hormones and neuropeptides precursors. Single nucleotide polymorphisms in this gene may increase susceptibility to myocardial infarction and type 2 diabetes. This gene may also play a role in tumor development and progression.
