MBS703495 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the phospholipase A2, group VII (platelet-activating factor acetylhydrolase, plasma) (PLA2G7) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PLA2G7. The ELISA analytical biochemical technique of the MBS703495 kit is based on PLA2G7 antibody-PLA2G7 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PLA2G7 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PLA2G7. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay||This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for Lp-PLA2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Lp-PLA2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Lp-PLA2 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Lp-PLA2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a secreted enzyme that catalyzes the degradation of platelet-activating factor to biologically inactive products. Defects in this gene are a cause of platelet-activating factor acetylhydrolase deficiency. Function: Modulates the action of platelet-activating factor (PAF) by hydrolyzing the sn-2 ester bond to yield the biologically inactive lyso-PAF. Has a specificity for substrates with a short residue at the sn-2 position. It is inactive against long-chain phospholipids.

Catalytic activity: 1-alkyl-2-acetyl-sn-glycero-3-phosphocholine + H2O = 1-alkyl-sn-glycero-3-phosphocholine + acetate.
