MBS135510 is a ready-to-use microwell, strip-or-full plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Multiple tPA total antigen assay (PLAT) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PLAT. The ELISA analytical biochemical technique of the MBS135510 kit is based on PLAT antibody-PLAT antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PLAT antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PLAT. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This human tissue-type plasminogen activator (tPA) total antigen assay is intended for the quantitative determination of total tissue plasminogen activator in human plasma and other biological fluids. For research use only.

Principle of the Assay: Human tPA will bind to the affinity purified capture antibody coated on the microtiter plate. Free, latent and complexed tPA will bind to the plate. After appropriate washing steps, monoclonal anti-human tPA primary antibody binds to the captured protein. Excess primary antibody is washed away and bound antibody is reacted with the peroxidase conjugated secondary antibody. Following an additional washing step, TMB is used for color development at 450nm. A standard calibration curve is prepared along with the samples to be measured using dilutions of human tPA. Color development is proportional to the concentration of tPA in the samples.

Background: tPA is a serine protease that catalyzes the activation of plasminogen to plasmin [1]. Clinical studies have indicated that high tPA levels may increase the risk for thrombosis [2], whereas decreased levels may cause neuronal plasticity and degeneration [3].

This gene encodes tissue-type plasminogen activator, a secreted serine protease which converts the proenzyme plasminogen to plasmin, a fibrinolytic enzyme. Tissue-type plasminogen activator is synthesized as a single chain which is cleaved by plasmin to a two chain disulfide linked protein. This enzyme plays a role in cell migration and tissue remodeling. Increased enzymatic activity causes hyperfibrinolysis, which manifests as excessive bleeding; decreased activity leads to hypofibrinolysis which can result in thrombosis or embolism. Alternative splicing of this gene results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]
