Other Notes
Small volumes of anti-HGS antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Applications Tested/Suitable for anti-HGS antibody
Peptide ELISA (EIA), Immunohistochemistry (IHC)
Application Notes for anti-HGS antibody
Peptide ELISA: Antibody detection limit dilution 1: 32000.
Immunohistochemistry: In paraffin embedded Human Testis shows strong staining of Leydig cells. Recommended concentration, 1-2ug/ml.
Western Blot: Preliminary experiments gave bands at approx 140kDa and 26kDa in Human lysates of monocytic leukemia cell line U937 after 0.3ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the bands we observe given the calculated size of 86.2kDa according to NP_004703.1. Both detected bands were successfully blocked by incubation with the immunizing peptide (and BLAST results with the immunizing peptide sequence did not identify any other proteins to explain the additional bands).

Immunohistochemistry (IHC) of anti-HGS antibody
(2.5ug/ml) staining of paraffin embedded Human Testis. Steamed antigen retrieval with citrate buffer pH 6, AP-staining.

NCBI Summary for HGS
The protein encoded by this gene regulates endosomal sorting and plays a critical role in the recycling and degradation of membrane receptors. The encoded protein sorts monoubiquitinated membrane proteins into the multivesicular body, targeting these proteins for lysosome-dependent degradation. [provided by RefSeq, Dec 2010]
UniProt Comments for HGS
Hrs: a zinc finger protein which is rapidly tyrosine phosphorylated in cells stimulated with various growth factors. Hrs mRNA is expressed in all the human adult and fetal tissues tested.

Protein type: Motility/polarity/chemotaxis; Adaptor/scaffold

Chromosomal Location of Human Ortholog: 17q25

Cellular Component: cytoplasm; cytosol; early endosome; early endosome membrane; endosome; intracellular membrane-bound organelle
