A synthetic peptide corresponding to a sequence at the C-terminus of human Phospholipase A2 (686-700aa PNQAFKRLHDLMHFN), different from the related rat and mouse sequences by one amino acid.
Each vial contains 5mg BSA, 0.9mg NaCl, 0.2mg Na2HPO4, 0.05mg Thimerosal, 0.05mg NaN3.
Add 0.2ml of distilled water will yield a concentration of 500ug/ml.
Preparation and Storage
At -20 degree C for one year. After reconstitution, at 4 degree C for one month. It can also be aliquotted and stored frozen at -20 degree C for a longer time. Avoid repeated freezing and thawing.

Background: PLA2G4A(Phospholipase A2, Group IVA), is an enzyme that in humans is encoded by the PLA2G4A gene. Tay et al.(1995) mapped the PLA2G4A gene to rat chromosome 13 by PCR-based intercross genotyping and to human 1q25 by fluorescence in situ hybridization. By site-directed mutagenesis and biochemical analysis of the recombinant protein, Sharp et al.(1994) determined that ser228 participates in the catalytic mechanism of cPLA2 and that both the phospholipase A2 and the lysophospholipase activities are catalyzed by the same active site residue(s). PLA2G4A, the cytosolic phospholipase A2, appears to subserve transmembrane signaling responses to extracellular ligands(Skorecki, 1995).

This gene encodes a member of the cytosolic phospholipase A2 group IV family. The enzyme catalyzes the hydrolysis of membrane phospholipids to release arachidonic acid which is subsequently metabolized into eicosanoids. Eicosanoids, including prostaglandins and leukotrienes, are lipid-based cellular hormones that regulate hemodynamics, inflammatory responses, and other intracellular pathways. The hydrolysis reaction also produces lysophospholipids that are converted into platelet-activating factor. The enzyme is activated by increased intracellular Ca(2+) levels and phosphorylation, resulting in its translocation from the cytosol and nucleus to perinuclear membrane vesicles. Selectively hydrolyzes arachidonyl phospholipids in the sn-2 position releasing arachidonic acid. Arachidonic acid is a precursor for the eicosanoids that are involved in hemodynamic regulation, inflammatory responses, and other cellular processes. Promotes cerebellar long-term depression and motor learning. Translocates to the Golgi and endoplasmic reticulum in a calcium-dependent fashion. Translocation and activation of at the ER facilitates the process of ER stress. Regulates the biogenesis of lipid droplets, a process dependent on JNK and ceramide kinase. Stimulated by agonists such as ATP, EGF, thrombin and bradykinin as well as by cytosolic Ca2+. The N-terminal C2 domain, by its association with lipid membranes, mediates the regulation of CPLA2 by presenting the active site to its substrate in response to elevations of cytosolic Ca2+.
