For Research Use Only. Not for use in diagnostic procedures.
Clonality
Polyclonal
Isotype
IgG
Host
Rabbit
Species Reactivity
Human (Expected from Sequence), Mouse (Expected from Sequence), Xenopus (Expected from Sequence)
Specificity
This item is specific for rat calcium/calmodulin kinase II (CaMK II) alpha and CaMK II beta, when phosphorylated at Thr286.

CaMKII is a multi-functional calcium and calmodulin-dependent kinase which has an important role in cell signalling and becomes autonomously active when phosphorylated at Thr286.

Purified
Purified IgG - liquid
Perservative Stabilisers
0.09% Sodium Azide
0.01% Bovine Serum Albumin
50% Glycerol
Immunogen
Synthetic phosphopeptide corresponding to an amino acid sequence within CaMK II which includes phosphorylated Thr286.
Buffer Solution
Antiserum Preparation
Antisera to phosphorylated rat CaMK II were raised by repeated immunisations of rabbit with highly purified antigen. Purified IgG was prepared by affinity chromatography.
Target Species
Rat
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. This product should be stored undiluted. Storage in frost free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of despatch.
Other Notes
Small volumes of anti-CaMK II antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

Application Notes for anti-CaMK II antibody
Western Blot: This item detects bands of approximately 50 and 60 kDa in rat brain lysates, corresponding to CaMKII alpha and CaMK II beta respectively.
Western Blotting: Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, threonine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.

UniProt Comments for CaMK II
CAMK2A: a protein kinase of the CAMK2 family. A prominent kinase in the central nervous system that may function in long-term potentiation and neurotransmitter release. Member of the NMDAR signaling complex in excitatory synapses that may regulate NMDAR-dependent potentiation of the AMPAR and synaptic plasticity. The holoenzyme is composed of four different chains: alpha, beta, gamma, and delta. The different chains assemble into homo- or heteromultimeric holoenzymes composed of 8 to 12 subunits. May interact with BAALC, MPDZ, SYN1 and synGAP. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Protein kinase, CAMK; EC 2.7.11.17; Protein kinase, Ser/Thr (non-receptor); Kinase, protein; CAMK group; CAMK2 family

Cellular Component: dendrite cytoplasm; presynaptic membrane; mitochondrion; cell soma; axon; postsynaptic density; dendrite; nucleus; cell junction

Molecular Function: calmodulin binding; protein serine/threonine kinase activity; protein binding; glutamate receptor binding; protein homodimerization activity; calmodulin-dependent protein kinase activity; GTPase activating protein binding; kinase activity; ATP binding
