The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PPP2R1A. The ELISA analytical biochemical technique of the MBS926939 kit is based on PPP2R1A antibody-PPP2R1A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PPP2R1A antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PPP2R1A. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay||This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PPP2R1A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PPP2R1A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PPP2R1A is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PPP2R1A bound in the initial step. The color development is stopped and the intensity of the color is measured.

Protein phosphatase 2 is one of the four major Ser/Thr phosphatases, and it is implicated in the negative control of cell growth and division. It consists of a common heteromeric core enzyme, which is composed of a catalytic subunit and a constant regulatory subunit, that associates with a variety of regulatory subunits. The constant regulatory subunit A serves as a scaffolding molecule to coordinate the assembly of the catalytic subunit and a variable regulatory B subunit. This gene encodes an alpha isoform of the constant regulatory subunit A. Alternatively spliced transcript variants have been described. [provided by RefSeq, Apr 2010]
