Related Product Information for anti-VTCN1 antibody

B7H4, also known as B7x, is a costimulatory protein which is reported to function as a negative regulator of T-cell mediated immunity. Although B7H4 binds an unknown receptor, it is thought to deliver an inhibitory signal to T-cells preventing their proliferation, cell cycle progression and interleukin-2 production. B7H4 deficient mice are only minimally affected; suggesting B7H4 is important in the fine tuning of the T-cell mediated immune response. B7H4 is expressed on activated T-cells, B-cells, monocytes and dendritic cells. Aberrant expression has been associated with cancers of the lung, breast and ovary in humans. B7H4 could be a useful prognostic marker in Renal Cell Carcinoma (RCC).

NCBI Summary for VTCN1    B7H4 belongs to the B7 family (see CD80; MIM 112203) of costimulatory proteins. These proteins are expressed on the surface of antigen-presenting cells and interact with ligands (e.g., CD28; MIM 186760) on T lymphocytes.[supplied by OMIM]

UniProt Comments for VTCN1    VTCN1: Negatively regulates T-cell-mediated immune response by inhibiting T-cell activation, proliferation, cytokine production and development of cytotoxicity. When expressed on the cell surface of tumor macrophages, plays an important role, together with regulatory T-cells (Treg), in the suppression of tumor- associated antigen-specific T-cell immunity. Involved in promoting epithelial cell transformation. Belongs to the immunoglobulin superfamily. BTN/MOG family. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, integral

Chromosomal Location of Human Ortholog: 1p13.1

Cellular Component: integral to membrane; external side of plasma membrane

Molecular Function: receptor binding

Biological Process: response to protozoan; immune system process; positive regulation of T cell proliferation; negative regulation of T cell activation

Research Articles on VTCN1    1. Novel role of B7-H4 molecule in the suppressive effect of hBMSCs on T-cell activation and proliferation.
2. B7-H4 directly promotes malignant transformation of ovarian cancer cell line.
3. B7-H4 gene polymorphism may contribute to the sporadic breast cancer risk and prognosis in Chinese Han women.
4. Observational study of gene-disease association. (HuGE Navigator)
5. Observational study and genome-wide association study of gene-disease association. (HuGE Navigator)
6. renal cell carcinoma patients are more likely to have detectable sB7x compared with controls and higher sB7x levels correlate with advanced tumor stage
7. B7-H4 was expressed more often in pancreatic ductal carcinoma than was p53.
8. B7-H4 expression was low in normal ovaries and in benign tumors while half of early stage and two-thirds of late stage cancers over-expressed B7-H4.
9. Tight regulation of B7-H4 expression at the translational level is observed in normal peripheral tissues and a potential role indicated in the evasion of tumor immunity.
10. patients with renal cell carcinoma (RCC) tumors expressing both B7-H4 and B7-H1 are at an even greater risk of death from RCC
