Related Product Information for anti-GABRB2 antibody

GABA (A) receptors are ligand-gated chloride channels that play a role as inhibitory neurotransmitters. They are known targets for certain classes of environmental and pharmaceutical compounds because a number of drugs interact with binding sites on GABA (A). Some of these drugs include benzodiazepines, anticonvulsants, and anesthetics.

 Applications Tested/Suitable for anti-GABRB2 antibody

Western Blot (WB), Immunoprecipitation (IP), Immunohistochemistry (IHC)

Application Notes for anti-GABRB2 antibody    Suitable for use in Immunohistochemistry, Western Blot and Immunoprecipitation.
Dilution: Western Blot: 0.5-2ug/ml detected the GABA A Receptor beta 2,3 subunits from 20ug of rat brain microsomal preparation. Rat brain microsomal preparation was resolved by electrophoresis, transferred to nitrocellulose and probed with G1016 (2ug/ml). Proteins were visualized using a IgG (HRP) GtxMo and ECL.
Immunoprecipitation: 4ug immunoprecipated the GABA A Receptor beta 2,3 subunits from 500ug of rat brain microsomal preparation. Due to interference of the IgG heavy chain, the immunoprecipitated GABA A reseptor complex was confirmed using G1015-06.
Immunohistochemistry: Detects GABA A Receptor beta 2,3 subunits in paraffin embedded and frozen rat brain sections fixed with 4% paraformaldehyde or 1% glutaraldehyde and permeabilized with Triton X-100.

NCBI Summary for GABRB2    The gamma-aminobutyric acid (GABA) A receptor is a multisubunit chloride channel that mediates the fastest inhibitory synaptic transmission in the central nervous system. This gene encodes GABA A receptor, beta 2 subunit. It is mapped to chromosome 5q34 in a cluster comprised of genes encoding alpha 1 and gamma 2 subunits of the GABA A receptor. Alternative splicing of this gene generates 2 transcript variants, differing by a 114 bp insertion. [provided by RefSeq]

UniProt Comments for GABRB2    Function: GABA, the major inhibitory neurotransmitter in the vertebrate brain, mediates neuronal inhibition by binding to the GABA/benzodiazepine receptor and opening an integral chloride channel.

Subunit structure: Generally pentameric. There are five types of GABA(A) receptor chains: alpha, beta, gamma, delta, and rho. Binds UBQLN1. Interacts with KCTD8, KCTD12 and KCTD16; this interaction determines the pharmacology and kinetics of the receptor response, the KCTD proteins markedly accelerating the GABA-B response, although to different extents

Subcellular location: Cell junction › synapse › postsynaptic cell membrane; Multi-pass membrane protein. Cell membrane; Multi-pass membrane protein.

Tissue specificity: Isoform 1 and isoform 2 show reduced expression in schizophrenic brain. Isoform 3 shows increased expression in schizophrenic and bipolar disorder brains while isoform 4 shows reduced expression. Ref.3 Ref.7

Sequence similarities: Belongs to the ligand-gated ion channel (TC 1.A.9) family. Gamma-aminobutyric acid receptor (TC 1.A.9.5) subfamily. GABRB2 sub-subfamily. [View classification]
