Mucosal vascular addressin-1 (MAdCAM-1) is a type I transmembrane glycoprotein expressed at high levels on high endothelial venules (HEV) of Peyer's patches and mesenteric lymph nodes, and on flatwalled venules within the gut lamina propria. It is also expressed on sinus-lining cells in the spleen.1-3 The countereceptor or "homing receptor" for MAdCAM-1 is alpha 4 beta 7 integrin (also known as LPAM-1). MAdCAM-1 is also a facultative ligand for CD62L/L-selectin.4, 5 The monoclonal antibody MECA-367 binds to the first domain of MAdCAM-1 and blocks MAdCAM-1-dependent binding in vitro and lymphocyte homing to Peyer's patch HEV in vivo.1, 2, 4

 Characterization    To ensure lot-to-lot consistency, each batch of monoclonal antibody is tested by flow cytometry to conform to characteristics of a standard reference reagent.

 Preparation and Storage    The purified (UNLB) antibody is supplied as 0.5 mg of purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.2. No preservatives or amine-containing buffer salts added. Store at 2- 8 degree C.
The fluorescein (FITC) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C.
The biotin (BIOT) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C.
The low endotoxin, azide-free (LE/AF) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of PBS. Aliquot and store at or below -20 degree C.
Protect conjugated forms from light. Aliquot and freeze the low endotoxin, azide-free product at -20 degree C immediately upon receipt. Each reagent is stable for the period shown on the bottle label if stored as directed.
 Other Notes    Small volumes of anti-MADCAM1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

 NCBI Summary for MADCAM1    The protein encoded by this gene is an endothelial cell adhesion molecule that interacts preferentially with the leukocyte beta7 integrin LPAM-1 (alpha4beta7), L-selectin, and VLA-4 (alpha4beta1) on myeloid cells to direct leukocytes into mucosal and inflamed tissues. It is a member of the immunoglobulin family and is similar to ICAM1 and VCAM1. At least seven alternatively spliced transcripts encoding different protein isoforms have been found for this gene, but the full-length nature of some variants has not been determined.  UniProt Comments for MADCAM1    Function: Cell adhesion leukocyte receptor expressed by mucosal venules, helps to direct lymphocyte traffic into mucosal tissues including the Peyer patches and the intestinal lamina propria. It can bind both integrin alpha-4/beta-7 and L-selectin, regulating both the passage and retention of leukocytes. Isoform 2, lacking the mucin-like domain, may be specialized in supporting integrin alpha-4/beta-7-dependent adhesion strengthening, independent of L-selectin binding.

Subcellular location: Membrane; Single-pass type I membrane protein.

Tissue specificity: Highly expressed on high endothelial venules (HEV) and lamina propia venules found in the small intestine, and to a lesser extent in the colon and spleen. Very low levels of expression found in pancreas and brain. Not expressed in the thymus, prostate, ovaries, testis, heart, placenta, lung, liver, skeletal muscle, kidney or peripheral blood leukocytes. Ref.1 Ref.2

Post-translational modification: The Ser/Thr-rich mucin-like domain may provide possible sites for O-glycosylation

Polymorphism: The number of repeats in the mucin domain varies between 5 and 8 repeats.
