 Preparation and Storage    May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Use on 30 um, free floating, 4% paraformaldehyde fixed rat brain tissue. All steps are performed under constant agitation.
1. 3 x 10 minute washes in TBS (with or without 0.25% Triton).
2. Incubate for 30 minutes in TBS with 3% serum (same as host from secondary antibody).
3. Incubate M9520-13B diluted appropriately in TBS with 1% serum (same as host from secondary antibody) (with or without 0.25% Triton) for 2 hours at RT followed by 16 hours at 4 degree C.
4. 3 x 10 minute washes in TBS.
5. Incubate with secondary antibody diluted appropriately in TBS with 1% serum (same as host from secondary antibody).
6. 3 x 10 minute washes in TBS.
7. ABC Elite (1:200) in TBS.
8. 2 x 10 minute washes in TBS.
9. 1 x 10 minute wash in phosphate buffer (no saline).
10. DAB reaction with 0.06% NiCl added for intensification.
11. 2 x 10 minute washes in PBS.
12. 1 x 10 minute wash in phosphate buffer (no saline).
 Other Notes    Small volumes of anti-CHRM2 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

 NCBI Summary for CHRM2    The muscarinic cholinergic receptors belong to a larger family of G protein-coupled receptors. The functional diversity of these receptors is defined by the binding of acetylcholine to these receptors and includes cellular responses such as adenylate cyclase inhibition, phosphoinositide degeneration, and potassium channel mediation. Muscarinic receptors influence many effects of acetylcholine in the central and peripheral nervous system. The muscarinic cholinergic receptor 2 is involved in mediation of bradycardia and a decrease in cardiac contractility. Multiple alternatively spliced transcript variants have been described for this gene. [provided by RefSeq]

 UniProt Comments for CHRM2    mAChR M2: The muscarinic acetylcholine receptor mediates various cellular responses, including inhibition of adenylate cyclase, breakdown of phosphoinositides and modulation of potassium channels through the action of G proteins. Primary transducing effect is adenylate cyclase inhibition. Genetic variations in CHRM2 can influence susceptibility to major depressive disorder (MDD). MDD is one of the most common psychiatric disorders. MDD is a complex trait characterized by one or more major depressive episodes without a history of manic, mixed, or hypomanic episodes. A major depressive episode is characterized by at least 2 weeks during which there is a new onset or clear worsening of either depressed mood or loss of interest or pleasure in nearly all activities. Four additional symptoms must also be present including changes in appetite, weight, sleep, and psychomotor activity; decreased energy; feelings of worthlessness or guilt; difficulty thinking, concentrating, or making decisions; or recurrent thoughts of death or suicidal ideation, plans, or attempts. The episode must be accompanied by distress or impairment in social, occupational, or other important areas of functioning. Belongs to the G-protein coupled receptor 1 family.
