Related Product Information for anti-Csk antibody

This monoclonal antibody recognizes an active form of c-Src which is not phosphorylated at tyrosine 529. Src (also known as pp60 src) is a non-receptor tyrosine kinase involved in signal transduction in many biological systems and implicated in the development of human tumors. There are two critical phosphorylation sites of tyrosine on Src, tyrosine 418 and tyrosine 529 (referring to human Src sequence). The tyrosine 418 is located in the catalytic domain and is one of the autophosphorylation sites. Full catalytic activity of Src requires phosphorylation of tyrosine 418. The tyrosine 529 is located near the carboxyl terminus of Src and acts as a negative regulator, in that Src is held in the inactive form through an intramolecular interaction between the SH2 domain and the carboxyl terminus when tyrosine 529 is phosphorylated by Csk. This conformation blocks phosphorylation of tyrosine 418 at the catalytic domain, thereby preventing Src activation. When tyrosine 529 is dephosphorylated, tyrosine 418 can be maximally phosphorylated and Src becomes active. This antibody also recognizes the other members of Src family including Fyn, Yes, and Fgr.

UniProt Comments for Csk    Function: Non-receptor tyrosine-protein kinase that plays an important role in the regulation of cell growth, differentiation, migration and immune response. Phosphorylates tyrosine residues located in the C-terminal tails of Src-family kinases (SFKs) including LCK, SRC, HCK, FYN, LYN or YES1. Upon tail phosphorylation, Src-family members engage in intramolecular interactions between the phosphotyrosine tail and the SH2 domain that result in an inactive conformation. To inhibit SFKs, CSK is recruited to the plasma membrane via binding to transmembrane proteins or adapter proteins located near the plasma membrane. Suppresses signaling by various surface receptors, including T-cell receptor (TCR) and B-cell receptor (BCR) by phosphorylating and maintaining inactive several positive effectors such as FYN or LCK

Catalytic activity: ATP + a [protein]-L-tyrosine = ADP + a [protein]-L-tyrosine phosphate.

Subunit structure: Homodimer (via SH3-domain). Interacts with PTPN8. Interacts with phosphorylated SIT1, PAG1, LIME1 and TGFB1I1; these interactions serve to recruit CSK to the membrane where it can phosphorylate and inhibit Src-family kinases. Interacts with SRCIN1. Interacts with RHOH. Interacts (via SH2 domain) with SCIMP

By similarity. Ref.5 Ref.6

Subcellular location: Cytoplasm

By similarity. Cell membrane

By similarity. Note: Mainly cytoplasmic, also present in lipid rafts

By similarity.

Tissue specificity: Enriched in lymphoid tissues.

Domain: The architecture of this protein is similar to that of Src-family kinases (SFKs) with one N-terminal SH3 domain, one SH2 domain, and a C-terminal kinase domain

By similarity.

Post-translational modification: Phosphorylated at Ser-364 by PKA, leading to increased activity. Autophosphorylated

By similarity.

Sequence similarities: Belongs to the protein kinase superfamily. Tyr protein kinase family. CSK subfamily.Contains 1 protein kinase domain.Contains 1 SH2 domain.Contains 1 SH3 domain.

Research Articles on Csk    1. Recognition of Src-family kinases (SFKs) by Csk is mainly governed by interactions between motifs located distally from the active site of Csk and determinants spatially separate from the carboxyl-terminal regulatory tyrosine residue in SFKs.
