Specificity    By immunohistochemistry, it specifically recognizes a protein in melanocytes and melanomas. This MAb reacts with junctional and blue nevus cells and variably with fetal and neonatal melanocytes. Intradermal nevi, normal adult melanocytes, and non-melanocytic cells are negative. It does not stain tumor cells of epithelial, lymphoid, glial, or mesenchymal origin. This Mab labels formalin-fixed, paraffin-embedded melanomas and other tumors showing melanocytic differentiation.

 Applications Tested/Suitable for anti-SILV antibody

ELISA (EIA), Flow Cytometry(FC/FACS, Immunofluorescence (IF), Immunohistology(IH)

Application Notes for anti-SILV antibody    ELISA (For coating, order Ab without BSA); Flow Cytometry (0.5-1ug/million cells); Immunofluorescence (1-2ug/ml); Immunohistology (Frozen & Formalin-fixed) (1-2ug/ml for 30 minutes at RT) (Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes) Optimal dilution for a specific application should be determined.

NCBI Summary for SILV    This gene encodes a melanocyte-specific type I transmembrane glycoprotein. The encoded protein is enriched in melanosomes, which are the melanin-producing organelles in melanocytes, and plays an essential role in the structural organization of premelanosomes. This protein is involved in generating internal matrix fibers that define the transition from Stage I to Stage II melanosomes. This protein undergoes a complex pattern of prosttranslational processing and modification that is essential to the proper functioning of the protein. A secreted form of this protein that is released by proteolytic ectodomain shedding may be used as a melanoma-specific serum marker. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Jan 2011]
Section Bar
UniProt Comments for SILV    SILV: Plays a central role in the biogenesis of melanosomes. Involved in the maturation of melanosomes from stage I to II. The transition from stage I melanosomes to stage II melanosomes involves an elongation of the vesicle, and the appearance within of distinct fibrillar structures. Release of the soluble form, ME20-S, could protect tumor cells from antibody mediated immunity. Belongs to the PMEL/NMB family. 5 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, integral; Vesicle

Chromosomal Location of Human Ortholog: 12q13-q14

Cellular Component: Golgi apparatus; endoplasmic reticulum membrane; melanosome; plasma membrane; extracellular region; integral to membrane

Molecular Function: protein binding

Biological Process: melanin biosynthetic process; melanosome organization and biogenesis
