Related Product Information for anti-MAP4K4 antibody

Nuclear factor kappa B (NF- kappa B) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NF- kappa B mediates the expression of a great variety of genes in response to extracellular stimuli including IL-1, TNF alpha , LPS, and mitogens. A serine/threonine protein kinase which mediates NF- kappa B activation by IL-1, TNF alpha , and CD95 has been identified and designated NIK (for NF- kappa B inducing kinase). NIK is an activator of I kappa B kinase alpha and beta (IKK alpha and IKK beta ). Therefore, NIK is a key molecule in the NF- kappa B signaling pathway leading to the expression of a variety of gene products in response to pro-inflammatory cytokines and bacterial products.

NCBI Summary for MAP4K4    The protein encoded by this gene is a member of the serine/threonine protein kinase family. This kinase has been shown to specifically activate MAPK8/JNK. The activation of MAPK8 by this kinase is found to be inhibited by the dominant-negative mutants of MAP3K7/TAK1, MAP2K4/MKK4, and MAP2K7/MKK7, which suggests that this kinase may function through the MAP3K7-MAP2K4-MAP2K7 kinase cascade, and mediate the TNF-alpha signaling pathway. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq]

UniProt Comments for MAP4K4    HGK: a Ser/Thr kinase of the STE20 subfamily. May play a role in the response to environmental stress and cytokines such as TNF-alpha. May interact with the SH3 domain of the adapter proteins Nck. Binds, via its CNH regulatory domain, to the N- terminal region of SPG3A. Upregulated in primary tumors and tumor cell lines. Required for anchorage-independent growth and ras-dependent focus formation in multiple cell lines. 5 isoforms of the human protein are produced by alternative splicing.

Protein type: Kinase, protein; Protein kinase, Ser/Thr (non-receptor); Protein kinase, STE; EC 2.7.11.1; STE group; STE20 family; MSN subfamily

Chromosomal Location of Human Ortholog: 2q11.2-q12

Cellular Component: cytoplasm

Molecular Function: protein serine/threonine kinase activity; protein binding; ATP binding; receptor signaling protein serine/threonine kinase activity

Biological Process: microvillus biogenesis; regulation of catalytic activity; regulation of JNK cascade; protein amino acid phosphorylation

Research Articles on MAP4K4    1. Data show that down-regulation of hepatocyte progenitor kinase-like kinase (HGK) can obviously inhibit the migration and invasion of HepG2 cells in vitro, and suggest that HGK may be a new therapeutic target for treatment of hepatocellular carcinoma.
