Antigen    Peptide from the extracellular loop 2 (EL2) region of human dopamine transporter (DAT), conjugated to keyhole limpet hemocyanin (KLH).
Immunogen Information    Synthetic peptide corresponding to amino acid residues from the extracellular loop 2 region conjugated to KLH
 Immunogen Species    Human
Species Reactivity Note    The antibody has been tested in Western blots of SDS-solubilized human striatal samples and in IHC applications with formaldehyde-fixed human and monkey brain sections.
Biological Significance    The dopamine transporter (DAT) is responsible for the reaccumulation of dopamine after it has been released. DAT antibodies and antibodies for other markers of catecholamine biosynthesis are widely used as markers for dopaminergic and noradrenergic neurons in a variety of applications including depression, schizophrenia, Parkinson's disease and drug abuse (Kish et al., 2001; Zhu et al., 2000; Zhu et al., 1999). Levels of DAT protein expression are altered by chronic drug administration (Wilson et al., 1996).

Preparation and Storage    For long term storage -20 degree C is recommended. Stable at -20 degree C for at least 1 year.
Other Notes    Small volumes of anti-SLC6A3 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for SLC6A3    This gene encodes a dopamine transporter which is a member of the sodium- and chloride-dependent neurotransmitter transporter family. The 3' UTR of this gene contains a 40 bp tandem repeat, referred to as a variable number tandem repeat or VNTR, which can be present in 3 to 11 copies. Variation in the number of repeats is associated with idiopathic epilepsy, attention-deficit hyperactivity disorder, dependence on alcohol and cocaine, susceptibility to Parkinson disease and protection against nicotine dependence.[provided by RefSeq, Nov 2009]

UniProt Comments for SLC6A3    Function: Amine transporter. Terminates the action of dopamine by its high affinity sodium-dependent reuptake into presynaptic terminals.

Subunit structure: Homooligomer; disulfide-linked. Interacts with PRKCABP and TGFB1I1. Interacts (via N-terminus) with SYNGR3 (via N-terminus)

By similarity. Interacts with SLC18A2

By similarity. Ref.13 Ref.14 Ref.15

Subcellular location: Membrane; Multi-pass membrane protein.

Involvement in disease: Parkinsonism-dystonia infantile (PKDYS) [MIM:613135]: A neurodegenerative disorder characterized by infantile onset of parkinsonism and dystonia. Other neurologic features include global developmental delay, bradykinesia and pyramidal tract signs.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.19

Miscellaneous: This protein is the target of psychomotor stimulants such as amphetamines or cocaine.

Sequence similarities: Belongs to the sodium:neurotransmitter symporter (SNF) (TC 2.A.22) family. SLC6A3 subfamily. [View classification]
