Immunohistochemistry (IHC) of anti-FOXO1 antibody    Immunohistochemical analysis of FOXO1 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

Immunofluorescence/Immunocytochemistry (IF/ICC) of anti-FOXO1 antibody    Immunofluorescent analysis of FOXO1 staining in MCF7 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 degree C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

NCBI Summary for FOXO1    This gene belongs to the forkhead family of transcription factors which are characterized by a distinct forkhead domain. The specific function of this gene has not yet been determined; however, it may play a role in myogenic growth and differentiation. Translocation of this gene with PAX3 has been associated with alveolar rhabdomyosarcoma. [provided by RefSeq, Jul 2008]
Section Bar
UniProt Comments for FOXO1    FOXO1A: a transcription factor of the forkhead family. A central regulator of metabolism in several cell types. May play an important role in myogenic growth and differentiation, and in the regulation of lipolysis in adipocytes by controlling the expression of adipose triglyceride lipase (ATGL), the rate-limiting lipolytic enzyme. Translocation of this gene with PAX3 has been associated with alveolar rhabdomyosarcoma. Contains 1 fork-head domain.

Research Articles on FOXO1    1. These findings suggest for the first time that FOXO1 is a promising candidate substrate for JNK, and the FOXO1-dependent DNA damage repair may be regulated positively by the JNK pathway in H1299 lung cancer cells.
