Antigen    Peptide representing a sequence that is specific for the gamma2-subunit of rat GABAA receptor. The sequence of the immunogen has no homology with any other GABAA R-subunit.
Immunogen Information    Synthetic peptide corresponding to amino acid residues specific to the gamma 2 subunit conjugated to KLH
 Immunogen Species    Rat
Reactivity Assumed Based on 100% Sequence Homology    Chicken, human, mouse, non-human primates and zebra fish
Species Reactivity Note    The antibody has been directly tested for reactivity in rat tissue. It is anticipated that the antibody will also work with bovine, chicken, human, mouse, non-human primate and zebra fish tissues based on the fact that these species have 100% homology with the amino acid sequence used as antigen.
Biological Significance    Gamma-aminobutyric acid (GABA) is the primary inhibitory neurotransmitter in the central nervous system, causing a hyperpolarization of the membrane through the opening of a Cl- channel associated with the GABAA receptor (GABAA-R) subtype. GABAA-Rs are important therapeutic targets for a range of sedative, anxiolytic, and hypnotic agents and are implicated in several diseases including epilepsy, anxiety, depression, and substance abuse. The GABAA-R is a multimeric subunit complex. To date six alphas, four betas and four gammas, plus alternative splicing variants of some of these subunits, have been identified (Olsen and Tobin, 1990; Whiting et al., 1999; Ogris et al., 2004). Injection in oocytes or mammalian cell lines of cRNA coding for alpha- and beta-subunits results in the expression of functional GABAA-Rs sensitive to GABA. However, co-expression of a gamma-subunit is required for benzodiazepine modulation. The various effects of the benzodiazepines in brain may also be mediated via different alpha-subunits of the receptor (McKernan et al., 2000; Mehta and Ticku, 1998; Ogris et al., 2004; Pöltl et al., 2003). Lastly, phosphorylation of beta-subunits of the receptor has been shown to modulate GABAA-R function (Brandon et al., 2003).

Preparation and Storage    For long term storage -20 degree C is recommended. Stable at -20 degree C for at least 1 year.
Other Notes    Small volumes of anti-GABRG2 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for GABRG2    subunit of GABA receptors, which bind benzodiazepines and act as inhibitory ion channels in the brain [RGD, Feb 2006]
Section Bar
UniProt Comments for GABRG2    Function: GABA, the major inhibitory neurotransmitter in the vertebrate brain, mediates neuronal inhibition by binding to the GABA/benzodiazepine receptor and opening an integral chloride channel.

Subunit structure: Generally pentameric. There are five types of GABA(A) receptor chains: alpha, beta, gamma, delta, and rho.

Subcellular location: Cell junction › synapse › postsynaptic cell membrane; Multi-pass membrane protein. Cell membrane; Multi-pass membrane protein.

Post-translational modification: Palmitoylated by ZDHHC3/GODZ; which may affect presynaptic clustering and/or cell surface stability

By similarity.

Miscellaneous: This subunit carries the benzodiazepine binding site.

Sequence similarities: Belongs to the ligand-gated ion channel (TC 1.A.9) family. Gamma-aminobutyric acid receptor (TC 1.A.9.5) subfamily. GABRG2 sub-subfamily. [View classification]
