 Species Reactivity Note    The antibody has been directly tested for reactivity in Western blots with rat tissue. It is anticipated that the antibody will react with bovine, canine, human, mouse, non-human primates and sheep based on the fact that these species have 100% homology with the amino acid sequence used as antigen.
 Biological Significance    Interferons are widely used therapeutic agents because of their anti tumor and antiviral effects and because of their modulatory effects on the immune system (Biron, 2001; Kirkwood, 2002). These cytokines produce their effects by binding to the Type 1 Interferon-alpha Receptor (IFNAR1). Down regulation of this receptor plays a key role in determining the magnitude and duration of cytokine signaling. This down regulation is thought to be influenced by phosphorylation of Serine 535 and 539 in the IFNAR1 (Kumar et al., 2003).

 Preparation and Storage    For long term storage -20 degree C is recommended. Stable at -20 degree C for at least 1 year.
 Other Notes    Small volumes of anti-IFNAR1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap.  NCBI Summary for IFNAR1    The protein encoded by this gene is a type I membrane protein that forms one of the two chains of a receptor for interferons alpha and beta. Binding and activation of the receptor stimulates Janus protein kinases, which in turn phosphorylate several proteins, including STAT1 and STAT2. The encoded protein also functions as an antiviral factor. [provided by RefSeq, Jul 2008]

 UniProt Comments for IFNAR1    Function: Associates with IFNAR2 to form the type I interferon receptor. Receptor for interferons alpha and beta. Binding to type I IFNs triggers tyrosine phosphorylation of a number of proteins including JAKs, TYK2, STAT proteins and IFNR alpha- and beta-subunits themselves.

Subunit structure: Heterodimer with IFNAR2; in presence of interferon alpha and beta ligands, the heterodimer forms the type I interferon receptor. Interacts with STAT1 and STAT2; the interaction requires its phosphorylation at Tyr-466. Interacts with IFNAR2. Ref.11 Ref.12

Subcellular location: Isoform 1: Membrane; Single-pass type I membrane protein.

Tissue specificity: IFN receptors are present in all tissues and even on the surface of most IFN-resistant cells. Isoform 1, isoform 2 and isoform 3 are expressed in the IFN-alpha sensitive myeloma cell line U266B1. Isoform 2 and isoform 3 are expressed in the IFN-alpha resistant myeloma cell line U266R. Isoform 1 is not expressed in IFN-alpha resistant myeloma cell line U266R. Ref.9

Post-translational modification: Phosphorylated on tyrosine residues by TYK2 tyrosine kinase. Ref.10Palmitoylation at Cys-463 is required for the activation of STAT1 and STAT2. Ref.14

Sequence similarities: Belongs to the type II cytokine receptor family.Contains 4 fibronectin type-III domains.

Sequence caution: The sequence AAH02590.1 differs from that shown. Reason: Contaminating sequence. Potential poly-A sequence.
