Specificity    This item specifically recognises the 66kDa serine/threonine kinase, PLK1 (polo-like kinase 1), when phosphorylated at Thr210.

PLK1 is a member of a group of polo-like kinases which play essential roles in the progression of mitosis. PLK1 has been implicated in various stages of mitosis, including promotion through the activation of cyclin B1/Cdk (mitosis promoting factor), the formation of functional mitotic bipolar spindles, and the maturation of centrosomes. The timing at which PLK1 phosphorylation occurs at Ser137 and Thr210 sites, may act as a regulator of PLK1 function through the mitotic cycle.

Preparation and Storage    Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of despatch.
Other Notes    Small volumes of anti-PLK1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

 Applications Tested/Suitable for anti-PLK1 antibody

Western Blot (WB)*

Application Notes for anti-PLK1 antibody    Western Blot: This item detects a band of approximately 66kDa in rat synaptic membrane cell lysates.
Western Blotting: Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, tyrosine phosphatase inhibitors such as 1mM Sodium Orthovanadate should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
