Preparation and Storage    Storage: Avoid repeated freeze/thaw cycles.
Store at 4C for frequent use.
Aliquot and store at -20C for 12 months.
Stability Test: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.

NCBI Summary for PAR4    This gene encodes a member of the protease-activated receptor subfamily, part of the G-protein coupled receptor 1 family of proteins. The encoded receptor is proteolytically processed to reveal an extracellular N-terminal tethered ligand that binds to and activates the receptor. This receptor plays a role in blood coagulation, inflammation and response to pain. Hypomethylation at this gene may be associated with lung cancer in human patients. [provided by RefSeq, Sep 2016]

UniProt Comments for PAR4    PAR4: a G-protein coupled receptor for activated thrombin or trypsin. Coupled to G proteins that stimulate phosphoinositide hydrolysis. Plays a role in platelets activation. Thrombin requires PAR4 for full spreading on a fibrinogen matrix. Widely expressed, with highest levels in lung, pancreas, thyroid, testis and small intestine. Not expressed in brain, kidney, spinal cord and peripheral blood leukocytes. Also detected in platelets. A proteolytic cleavage generates a new N-terminus that functions as a tethered ligand.

Protein type: GPCR, family 1; Membrane protein, integral; Membrane protein, multi-pass; Receptor, GPCR

Chromosomal Location of Human Ortholog: 19p13.11

Cellular Component: extracellular region; integral component of plasma membrane; plasma membrane

Molecular Function: G-protein coupled receptor activity; thrombin receptor activity

Biological Process: blood coagulation; elevation of cytosolic calcium ion concentration during G-protein signaling, coupled to IP3 second messenger (phospholipase C activating); phospholipase C-activating G-protein coupled receptor signaling pathway; platelet activation; platelet dense granule organization and biogenesis; positive regulation of release of sequestered calcium ion into cytosol; positive regulation of Rho protein signal transduction; response to wounding; signal transduction

Research Articles on PAR4    1. Prospective study demonstrated that AHRR and F2RL3 methylation levels had inverse relationships with self-reported smoking status and accurately discriminated for both current- and former- smoking. Moreover, methylation markers distinguished former smokers from never-smokers with high accuracy and significantly associated with an increased risk of lung cancer.
