Related Product Information for anti-CBL antibody

C-Cbl proto-oncogene is a ubiquitously expressed cytoplasmic adaptor protein that is especially predominant in hematopoietic cells (1,2). c-Cbl is rapidly tyrosine phosphorylated in response to stimulation of a variety of cell-surface receptors and becomes associated with a number of intracellular signaling molecules such as protein tyrosine kinases, phosphatidylinositol 3 kinase, Crk and 14-3-3 proteins (3,4). c-Cbl possesses a highly conserved N-terminal phosphotyrosine binding domain (TKB), a C3HC4 RING finger motif. TKB recognizes phosphorylated tyrosines on activated receptor tyrosine kinases (RTKs) as well as other nonreceptor tyrosine kinases. Its RING finger domain recruits ubiquitin-conjugating enzymes. These two domains are primarily responsible for c-Cbl proteins functioning as ubiquitin protein ligases and downregulators of RTKs (3). c-Cbl protein in human cancer tissues is frequently tyrosine phosphorylated in a tumor-specific manner (5). Phosphorylation of Tyr731 of c-Cbl provides a docking site for downstream signaling components such as p85 and Fyn (6).

NCBI Summary for CBL    The cbl oncogene was first identified as part of a transforming retrovirus which induces mouse pre-B and pro-B cell lymphomas. As an adaptor protein for receptor protein-tyrosine kinases, it positively regulates receptor protein-tyrosine kinase ubiquitination in a manner dependent upon its variant SH2 and RING finger domains. Ubiquitination of receptor protein-tyrosine kinases terminates signaling by marking active receptors for degradation. [provided by RefSeq]
Section Bar
UniProt Comments for CBL    CBL: an adapter protein that functions as a negative regulator of many signaling pathways that start from receptors at the cell surface. Acts as an E3 ubiquitin-protein ligase, which accepts ubiquitin from specific E2 ubiquitin-conjugating enzymes, and then transfers it to substrates promoting their degradation by the proteasome. Recognizes activated receptor tyrosine kinases, including PDGFA, EGF and CSF1, and terminates signaling. Participates in signal transduction in hematopoietic cells.

Research Articles on CBL    1. In human acute myeloid leukaemias, c-Cbl deletions are not common in the Australian population but they do raise the possibility that they might contribute to the pathogenesis.
