The protein encoded by this gene is a histone methyltransferase that methylates lysine-79 of histone H3. It is inactive against free core histones, but shows significant histone methyltransferase activity against nucleosomes. [provided by RefSeq, Aug 2011]

DOT1L: a protein-lysine N-methyltransferase of the seven beta-strand class. Catalyzes the mono-, di- and tri-methylation of H3K79. Methylated H3K79 is associated with active gene transcription. DOT1L activity drives proliferation of mixed lineage leukemia 1 protein (MLL)-rearranged leukemias, inducing the aberrant transcription of a number of leukemogenic genes including HOXA9 and MEIS1. Binds to DNA. Belongs to the class I-like SAM-binding methyltransferase superfamily. DOT1 family. 2 isoforms of the human protein are produced by alternative splicing.
