   Priciple of the assay: ADAMTS17 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ADAMTS17 antibody and an ADAMTS17-HRP conjugate. The assay sample and buffer are incubated together with ADAMTS17-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADAMTS17 concentration since ADAMTS17 from samples and ADAMTS17-HRP conjugate compete for the anti-ADAMTS17 antibody binding site. Since the number of sites is limited, as more sites are occupied by ADAMTS17 from the sample, fewer sites are left to bind ADAMTS17-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADAMTS17 concentration in each sample is interpolated from this standard curve.

 NCBI Summary for ADAMTS17    This gene encodes a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) protein family. ADAMTS family members share several distinct protein modules, including a propeptide region, a metalloproteinase domain, a disintegrin-like domain, and a thrombospondin type 1 (TS) motif. Individual members of this family differ in the number of C-terminal TS motifs, and some have unique C-terminal domains. The protein encoded by this gene has a high sequence similarity to the protein encoded by ADAMTS19, another family member. The function of this protein has not been determined.
