Product Description specifically for ATP5D elisa kit

   Intended Uses: This ATP5D ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine ATP5D. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: ATP5D ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ATP5D antibody and an ATP5D-HRP conjugate. The assay sample and buffer are incubated together with ATP5D-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ATP5D concentration since ATP5D from samples and ATP5D-HRP conjugate compete for the anti-ATP5D antibody binding site. Since the number of sites is limited, as more sites are occupied by ATP5D from the sample, fewer sites are left to bind ATP5D-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ATP5D concentration in each sample is interpolated from this standard curve.

 NCBI Summary for ATP5D    This gene encodes a subunit of mitochondrial ATP synthase. Mitochondrial ATP synthase catalyzes ATP synthesis, utilizing an electrochemical gradient of protons across the inner membrane during oxidative phosphorylation. ATP synthase is composed of two linked multi-subunit complexes: the soluble catalytic core, F1, and the membrane-spanning component, Fo, comprising the proton channel. The catalytic portion of mitochondrial ATP synthase consists of 5 different subunits (alpha, beta, gamma, delta, and epsilon) assembled with a stoichiometry of 3 alpha, 3 beta, and a single representative of the other 3. The proton channel consists of three main subunits (a, b, c). This gene encodes the delta subunit of the catalytic core.  UniProt Comments for ATP5D    ATP5D: Mitochondrial membrane ATP synthase (F(1)F(0) ATP synthase or Complex V) produces ATP from ADP in the presence of a proton gradient across the membrane which is generated by electron transport complexes of the respiratory chain. F-type ATPases consist of two structural domains, F(1) - containing the extramembraneous catalytic core, and F(0) - containing the membrane proton channel, linked together by a central stalk and a peripheral stalk. During catalysis, ATP turnover in the catalytic domain of F(1) is coupled via a rotary mechanism of the central stalk subunits to proton translocation. Part of the complex F(1) domain and of the central stalk which is part of the complex rotary element.
